兔骨髓間充質干細胞向胰島細胞分化的研究

【摘要】:旨在探討體外誘導兔骨髓間充質干細胞向胰島細胞分化的可行性,本研究采用二甲基亞砜聯(lián)合高濃度葡萄糖對分離純化的第3代兔BMSCs進行體外誘導,觀察誘導前后細胞形態(tài)學變化,用雙硫腙染色法鑒定胰島細胞,用免疫細胞化學法檢測胰島素的表達,用ELISA法檢測分泌到培養(yǎng)基中的胰島素含量。結果,試驗組在誘導7d出現(xiàn)細胞團樣集落,10d獲得排列緊密的細胞團,而對照組始終未見細胞團;經雙硫腙染色試驗組、免疫細胞化學染色試驗組均顯示陽性,對照組為陰性,ELISA檢測試驗組7和10d的胰島素含量分別為(27.82±0.43)和(27.89±0.25)mU.L-1,與對照組的(23.39±0.20)和(25.71±0.27)mU.L-1比較,差異極顯著(P0.01)。結果表明,二甲基亞砜聯(lián)合高糖能夠在較短的周期內誘導出胰島細胞。
[Abstract]:To explore the feasibility of inducing rabbit bone marrow mesenchymal stem cells to differentiate into islet cells in vitro, dimethyl sulfoxide (DMSO) combined with high concentration glucose was used to induce the third generation rabbit BMSCs in vitro. The changes of cell morphology were observed before and after induction. Islet cells were identified by dithizone staining, insulin expression was detected by immunocytochemistry and insulin content was detected by ELISA method. After 7 days of induction, the cells in the experimental group were closely arranged, but the cells in the control group were not seen at all, and the immunocytochemical staining group were all positive after dithizone staining. The insulin contents in the control group were 27.82 鹵0.43) and 27.89 鹵0.25 mU.L-1, respectively, which were significantly higher than those in the control group (23.39 鹵0.20) and the control group (25.71 鹵0.27)mU.L-1). The results showed that dimethyl sulfoxide combined with high glucose could induce islet cells in a short period of time.
