姜黃素殼聚糖微球制備及對2型糖尿病大鼠模型糖脂代謝的影響

【摘要】:目的制備姜黃素殼聚糖微球并研究其對2型糖尿病模型大鼠糖脂代謝的影響。方法用乳化分散-戊二醛交聯(lián)法制備姜黃素殼聚糖微球。高脂飼料連續(xù)喂養(yǎng)SD大鼠4周后,腹腔注射鏈脲佐菌素(25 mg·kg~(-1)),建立2型糖尿病大鼠模型。將大鼠隨機分為正常組、模型組和3個劑量實驗組(姜黃素殼聚糖微球,50,150,250 mg·kg~(-1))共5個組。于給藥前后,測定空腹血糖、空腹胰島素、餐后2 h血糖等指標,以及測定血清中總膽固醇(TC)、三酰甘油(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)含量;并用熒光定量PCR技術(shù)檢測血清中7個microRNA分子的表達量。結(jié)果制備的微球粒徑均勻,載藥量合適。給藥后,與正常組比較,模型組、實驗組大鼠的FBG、2PBG含量明顯升高;血清中TC、TG、LDL水平均明顯增高,HDL水平下降;microRNA的表達量也明顯上調(diào)(均P0.05)。高劑量實驗組組的療效更佳。結(jié)論姜黃素殼聚糖微球的制備簡單方便,重復性好,其可改善2型糖尿病大鼠中的糖脂代謝紊亂的狀況。
[Abstract]:Objective to prepare curcumin chitosan microspheres and study the effect of curcumin chitosan microspheres on glycolipid metabolism in type 2 diabetic rats. Methods curcumin chitosan microspheres were prepared by emulsification dispersion and glutaraldehyde crosslinking. The rat model of type 2 diabetes mellitus was established by intraperitoneal injection of streptozotocin 25 mg 路kg ~ (-1). The rats were randomly divided into normal group, model group and three experimental groups (curcumin chitosan microspheres 5050,250mg 路kg ~ (-1)). Fasting blood glucose was measured before and after administration of streptozotocin. Fasting insulin, 2 h postprandial blood glucose, and serum total cholesterol TCN, triglyceride TGN, high density lipoprotein (HDL), low density lipoprotein (LDL) were measured. Fluorescence quantitative PCR technique was used to detect the expression of seven microRNA molecules in serum. Results the microspheres prepared were of uniform size and suitable drug loading. Compared with the normal group, the FBG-2PBG content in the model group and the experimental group was significantly higher than that in the normal group. The level of TCU TGN LDL in serum was significantly increased and the level of HDL decreased. The expression of microRNA was also upregulated (P 0.05). Conclusion the preparation of curcumin chitosan microspheres is simple, convenient and reproducible. It can improve the disorder of glucose and lipid metabolism in type 2 diabetic rats.
