【摘要】：目的:比較2,4,6-三硝基苯磺酸(TNBS)和葡聚糖硫酸鈉(DSS)誘導(dǎo)的兩種腸炎大鼠模型。方法:SD大鼠隨機(jī)分為正常對照組、TNBS模型組和DSS模型組,每組10只。TNBS模型組大鼠采用TNBS 50%乙醇溶液以100 mg/kg一次性灌腸法建立腸炎模型,DSS模型組大鼠采用4%DSS水溶液連續(xù)7 d自由飲用建立腸炎模型。觀察各組大鼠一般情況,檢測血清總膽固醇(TC)和三酰甘油(TG)含量、結(jié)腸長度及質(zhì)量、結(jié)腸組織中髓過氧化物酶(MPO)活性,肉眼觀察結(jié)腸大體外觀形態(tài),蘇木精-伊紅染色觀察結(jié)腸病理學(xué)變化。結(jié)果:與正常對照組比較,TNBS模型組大鼠體質(zhì)量明顯減輕,結(jié)腸長度明顯縮短、單位長度質(zhì)量明顯增加,糞便性狀評分、血清TG含量、MPO活性、結(jié)腸大體外觀評分和病理學(xué)評分均明顯增加(P0.05或P0.01);DSS模型組大鼠體質(zhì)量明顯減輕,結(jié)腸長度明顯縮短,糞便性狀和結(jié)腸病理學(xué)評分均明顯增加(P0.05或P0.01)。與DSS模型組比較,TNBS模型組大鼠結(jié)腸長度明顯縮短、單位長度質(zhì)量明顯增加,血清TC、TG含量和結(jié)腸大體外觀評分、病理學(xué)評分明顯增加(P0.05或P0.01),組織病理學(xué)檢查可見明顯腸道潰瘍與粘連。結(jié)論:DSS誘導(dǎo)腸炎模型大鼠糞便評分較TNBS誘導(dǎo)模型低,TNBS誘導(dǎo)腸炎模型大鼠在腸道組織損傷方面較DSS誘導(dǎo)模型更嚴(yán)重、恢復(fù)更慢。
[Abstract]:Objective: to compare two kinds of enteritis rat models induced by TNBS) and sodium dextran sulfate (DSS). Methods: the rats were randomly divided into two groups: normal control group and DSS model group. TNBS 50% ethanol solution was used to establish enteritis model rats in each group of 10 rats. The rats in the model group were fed with 4DSS aqueous solution for 7 days to establish enteritis model. The general situation of the rats in each group was observed. The contents of total cholesterol (TC) and triglyceride (TG), the length and quality of colon, the activity of myeloperoxidase (MPO) in colon tissue, and the gross appearance of colon were observed with naked eyes. Results: compared with the normal control group, the body weight, colon length, unit length and fecal traits of TNBS model rats were significantly reduced, and the weight of the unit length was significantly increased, compared with the normal control group, the pathological changes of colon were observed by hematoxylin and eosin staining. Serum TG content and MPO activity, colonic gross appearance score and pathological score were significantly increased in DSS model group (P0.05 or P0.01). Compared with the DSS model group, the colonic length, the weight per unit length, the serum TCTG content and the colonic gross appearance score were significantly increased in the DSS model group. Histopathological examination showed obvious intestinal ulcer and adhesion. Conclusion the stool score of rats with TNBS induced enteritis is lower than that of TNBS-induced enteritis model rats. Surface is more serious than DSS model. Recovery is slower.
