基于COI基因片段的擬釘螺種類(lèi)鑒定初探

【摘要】:目的探討COI基因片段在擬釘螺種類(lèi)鑒定中的作用。方法收集來(lái)自福建省內(nèi)多個(gè)地區(qū)的疑似擬釘螺標(biāo)本,用形態(tài)學(xué)方法進(jìn)行物種鑒定后,取出螺肉提取基因組DNA,通過(guò)PCR的方式擴(kuò)增COI基因序列片段并測(cè)序,通過(guò)序列比對(duì)及系統(tǒng)進(jìn)化分析鑒定其物種。結(jié)果共采集到標(biāo)本13份,通過(guò)形態(tài)學(xué)方法將樣本鑒定為福建境擬釘螺屬。所有標(biāo)本經(jīng)實(shí)驗(yàn)擴(kuò)增到長(zhǎng)度為515~598bp的COI基因片段。13份標(biāo)本中11株的COI基因片段與γ擬釘螺屬最為相似(88.96%~97.82%),另外2株分別與擬釘螺屬下的武鳴擬釘螺(87.08%)及新擬釘螺屬下的開(kāi)放新擬釘螺(88.55%)最為相似。在屬水平上,13份標(biāo)本中僅有1份的形態(tài)學(xué)鑒定結(jié)果與基因鑒定結(jié)果吻合,其余12份樣本的形態(tài)學(xué)鑒定結(jié)果均與基因鑒定不同,兩種方法的鑒定結(jié)果存在較大差異。結(jié)論通過(guò)COI基因檢測(cè)可快速對(duì)擬釘螺亞科作出準(zhǔn)確判斷,但對(duì)屬水平的鑒定的準(zhǔn)確性還需要進(jìn)一步的研究。
[Abstract]:Objective to investigate the role of COI gene fragment in the species identification of Oncomelania hupensis. Genomic DNA was extracted from snail meat, COI gene sequence was amplified by PCR and sequenced. The species was identified by sequence alignment and phylogenetic analysis. The samples were identified as the genus Oncomelania hupensis by morphological method. The COI gene fragments of 11 of the 13 samples of 515598 BP in length were amplified by experiments. The COI gene fragment of 11 strains was the most similar to that of the genus 緯 Oncomelania, and the other 2 strains were subdivided into two strains. The results of morphological identification of 13 samples of Oncomelania hupensis were consistent with the results of gene identification, which were similar to those of Oncomelania hupensis (87.08) and the open Oncomelania hupensis (88.55) of the new Oncomelania hupensis. The results of morphological identification of the other 12 samples were different from those of gene identification, and there were great differences between the two methods. Conclusion the subfamilies of Oncomelania hupensis can be judged accurately by COI gene detection. However, the accuracy of generic identification needs further study.
