腎陽(yáng)虛外感小鼠模型建立及麻黃細(xì)辛附子湯干預(yù)研究

【摘要】:本論文首先對(duì)流感病毒致病特點(diǎn)以及中西醫(yī)防治流行性感冒(簡(jiǎn)稱流感)研究進(jìn)展進(jìn)行總結(jié),針對(duì)目前流感防治領(lǐng)域的重點(diǎn)和難點(diǎn)—流感高危人群的防治的重要性進(jìn)行了專門(mén)論述,隨后闡述了流感的高危人群多是屬于中醫(yī)的腎陽(yáng)虛體質(zhì),并以《傷寒雜病論》經(jīng)方為著眼點(diǎn),總結(jié)了麻黃細(xì)辛附子湯治療腎陽(yáng)虛外感的古今臨床應(yīng)用。在此基礎(chǔ)上進(jìn)行了實(shí)驗(yàn)研究,本課題首次建立了腎陽(yáng)虛外感病證動(dòng)物模型,并用麻黃細(xì)辛附子湯對(duì)其干預(yù),初步探討了麻黃細(xì)辛附子湯防治該病證的作用機(jī)理。 目的:通過(guò)病證結(jié)合的辦法,采用復(fù)合手段造模,建立了腎陽(yáng)虛外感動(dòng)物模型,利用該模型,觀察麻黃細(xì)辛附子湯對(duì)病證模型的影響,初步探討麻黃細(xì)辛附子湯干預(yù)腎陽(yáng)虛外感的作用機(jī)制。 方法:采用雌二醇腹腔注射復(fù)制腎陽(yáng)虛小鼠模型,在此基礎(chǔ)上鼻腔滴注流感病毒FM1建立了腎陽(yáng)虛外感病證模型,應(yīng)用麻黃細(xì)辛附子湯對(duì)該模型進(jìn)行干預(yù),比較動(dòng)物一般狀況、生理狀態(tài)、肺指數(shù)、死亡率等評(píng)價(jià)麻黃細(xì)辛附子湯對(duì)腎陽(yáng)虛外感模型的防治作用,并從細(xì)胞因子TNF-α、IFN-γ和抗氧化能力SOD、MDA水平初步探討麻黃細(xì)辛附子湯防治腎陽(yáng)虛外感作用機(jī)理。 結(jié)果:1.通過(guò)對(duì)小鼠一般狀況、生理狀態(tài)、肺指數(shù)、肺組織病理切片等觀察,表明本實(shí)驗(yàn)成功的建立了腎陽(yáng)虛外感病證模型。模型小鼠精神狀態(tài)低迷、趾溫、肛溫顯著性降低,死亡率升高,肺組織病理改變明顯。2.麻黃細(xì)辛附子湯干預(yù)后可明顯改善病證模型動(dòng)物的生理狀況,顯著性升高體溫,降低死亡率,TNF-α水平顯著性升高,肺組織免疫病理?yè)p傷降低。3.麻黃細(xì)辛附子湯干預(yù)后可明顯增加動(dòng)物的抗氧化能力,升高動(dòng)物血清當(dāng)中的SOD含量,降低MDA水平。 結(jié)論:雌二醇腹腔注射結(jié)合流感病毒FM1滴注可成功的建立腎陽(yáng)虛外感小鼠病證模型,,麻黃細(xì)辛附子湯可明顯改善腎陽(yáng)虛外感動(dòng)物的生理狀況,降低肺組織的病理?yè)p傷,顯著性升高血清中TNF-α水平,增強(qiáng)機(jī)體的抗氧化能力,從而達(dá)到對(duì)該病證的防治作用。
[Abstract]:This paper first summarizes the pathogenic characteristics of influenza virus and the progress in the prevention and treatment of influenza by traditional Chinese and western medicine. The importance of prevention and treatment of high risk groups of influenza in the field of influenza prevention and treatment at present is specially discussed. Then, it is expounded that most of the high-risk groups of influenza belong to the kidney-yang deficiency constitution of traditional Chinese medicine. The clinical application of ephedra Asarum aconite decoction in the treatment of kidney yang deficiency was summarized. Based on the experimental study, the animal model of kidney yang deficiency syndrome was established for the first time. The mechanism of ephedra Asarum aconite decoction in preventing and treating the disease syndrome was discussed. Objective: to establish an animal model of kidney yang deficiency by combining disease with syndromes, and to observe the effect of ephedra Asarum aconite decoction on the model of disease syndrome. Objective: to explore the mechanism of ephedra Asarum aconite decoction in the treatment of kidney yang deficiency. Methods: the mouse model of kidney yang deficiency was induced by intraperitoneal injection of estradiol, and the model of syndrome of kidney yang deficiency was established by nasal drip of influenza virus FM1. The model was treated with ephedra Asarum aconite decoction, and the general condition of animals was compared. Physiological status, lung index and mortality were evaluated to evaluate the preventive and therapeutic effects of ephedra Asarum aconite decoction on the exogenous model of kidney yang deficiency, and the mechanism of ephedra Asarum aconite decoction in preventing and treating kidney yang deficiency was preliminarily discussed from the levels of cytokine TNF- 偽, IFN- 緯 and antioxidant capacity (SODN MDA). Results 1. By observing the general condition, physiological state, lung index and pathological section of lung tissue in mice, the experimental results showed that the model of kidney yang deficiency was successfully established. The mental state of the model mice was low, the toe temperature and anal temperature were significantly decreased. After intervention of ephedrine Asarum aconite decoction, the physiological status of the model animals was improved, the body temperature was significantly increased, and the death rate was significantly decreased, and the level of TNF- 偽 was significantly increased. The immune pathological damage of lung tissue was decreased .3.After the intervention of Asarum ephedra decoction, the antioxidant ability of animals was increased, the content of SOD in serum was increased, and the level of MDA was decreased. Conclusion: intraperitoneal injection of estradiol combined with FM1 drip of influenza virus can successfully establish the model of kidney yang deficiency and exogenous disease in mice. Ephedra Asarum aconite decoction can obviously improve the physiological condition and reduce the pathological injury of lung tissue in the exogenous animals with deficiency of kidney yang. The level of TNF- 偽 in serum was significantly increased, and the antioxidant capacity of the body was enhanced, so that the prevention and treatment of the disease could be achieved.
