化痰類中藥干預肺纖維化模型大鼠肺組織中Nrf2含量的研究

【摘要】:目的探討部分化痰類藥物在博萊霉素所致模型大鼠肺纖維化中的抗氧化作用及途徑。方法采用氣管內(nèi)滴入博萊霉素的方法形成大鼠肺纖維化模型,以天南星、川貝母、海藻、胖大海、清半夏、白前、黃藥子、前胡等8味中藥的水煎液或吡非尼酮對模型大鼠進行灌胃干預,檢測各組大鼠肺組織中Nrf2的含量變化。結(jié)果 8味中藥組、對照組(模型組)、吡非尼酮組大鼠肺組織中的Nrf2含量明顯高于空白組(P0.05),川貝組、半夏組、天南星組、海藻組、胖大海組與對照組比較均P0.01,而吡非尼酮組與對照組比較P0.05,差異具有統(tǒng)計學意義。黃藥子組、前胡組、白前組等3味中藥組與對照組比較均P0.05。之后再將剩余的5味中藥組(川貝組、半夏組、天南星組、海藻組、胖大海組)與吡非尼酮組相比較均P0.05。說明上述5味中藥在改善Nrf2抗氧化指標及調(diào)控抗氧化基因表達方面與吡非尼酮作用相當。結(jié)論部分化痰類中藥能有效提高模型大鼠肺組織中Nrf2的含量,具有較強的修復細胞及機體解毒能力,其中川貝、半夏、天南星、海藻、胖大海的作用較為突出,證實了部分化痰類中藥可以通過干預機體的氧化-抗氧化機制對肺纖維化起到干預作用。
[Abstract]:Objective to investigate the antioxidation effect of some phlegm drugs on pulmonary fibrosis induced by bleomycin in rats. Methods Pulmonary fibrosis model of rats was established by intratracheal instillation of bleomycin. The model rats were treated with the decoction of eight kinds of traditional Chinese medicine, such as fat sea, clear Pinellia ternata, prewhiten, Huangzi, Qianhu, or pifenidone. The changes of Nrf2 content in lung tissue of rats in each group were detected. Results eight kinds of traditional Chinese medicine group were treated with traditional Chinese medicine. The content of Nrf2 in the control group (model group, pifenidone group) was significantly higher than that in the blank group (P 0.05), Chuanbei group, Pinellia ternata group, Tiannan Xing group, and seaweed group. Compared with the control group, the P0.01in the fat sea group, and the P0.05 in the pifenidone group compared with the control group, the difference was statistically significant. Compared with the control group, P0.05. the remaining five Chinese medicine groups (Chuanbei group, Pinellia ternata group, Tiannan Xing group, seaweed group) were added. Compared with pifenidone group, the P0.05. the results show that the above five herbs have the same effect in improving the antioxidant index of Nrf2 and regulating the expression of antioxidant gene. Conclusion the Chinese herbal medicine of partial phlegm reduction can effectively improve the model. The content of Nrf2 in lung tissue of rats. Has strong repair cells and body detoxification ability, among them Sichuan shellfish, Pinellia ternata, Tiannan star, seaweed, fat sea more prominent role, It is proved that partial phlegm can interfere with pulmonary fibrosis by intervening the oxidation-anti-oxidation mechanism.
