甘草對高脂血癥大鼠模型洛伐他汀藥代動(dòng)力學(xué)的影響

【摘要】:目的研究高脂血癥大鼠模型中甘草對洛伐他汀藥代動(dòng)力學(xué)過程的影響。方法將18只SD大鼠隨機(jī)分為對照組(n=6)和實(shí)驗(yàn)組(n=12)。實(shí)驗(yàn)組采用高脂飼料喂養(yǎng)大鼠,構(gòu)建高脂血癥大鼠模型,隨機(jī)分為洛伐他汀單獨(dú)給藥組(n=6)和洛伐他汀與甘草聯(lián)合給藥組(n=6)。聯(lián)合給藥組給予甘草顆粒,單獨(dú)給藥組給予相應(yīng)體積的生理鹽水,連續(xù)7 d后,2組大鼠單劑量灌胃給予洛伐他汀膠囊(20 mg/kg,0.5%CMC-Na溶液)。分別于給藥前和給藥后不同時(shí)間點(diǎn)采集血漿樣品,采用液相色譜-質(zhì)譜聯(lián)用方法測定血藥濃度,計(jì)算藥代動(dòng)力學(xué)參數(shù),并進(jìn)行組間比較。結(jié)果長期給予甘草導(dǎo)致洛伐他汀代謝產(chǎn)物洛伐他汀酸在高脂血癥大鼠中血漿暴露水平顯著升高,聯(lián)合給藥組平均C_(max)較單獨(dú)給藥組升高約80%(P0.05),而AUC0-t和AUC0-∞分別升高115%和109%(P=0.005和P=0.027)。聯(lián)合給藥組與單獨(dú)給藥組比較,洛伐他汀的C_(max)和AUC增加,但無統(tǒng)計(jì)學(xué)差異(P0.05)。結(jié)論甘草可抑制洛伐他汀在高脂血癥大鼠體內(nèi)的代謝,增加其體內(nèi)暴露水平。
[Abstract]:Objective to study the effect of licorice on the pharmacokinetic process of lovastatin in hyperlipidemic rats. Methods Eighteen SD rats were randomly divided into two groups: control group (n = 18) and experimental group (n = 12). The experimental group was fed with hyperlipidemic diet to establish a hyperlipidemic rat model. The rats were randomly divided into two groups: lovastatin group (n = 6) and lovastatin / licorice group (n = 6). The combination group was given licorice granules, and the control group was given a corresponding volume of normal saline. After 7 days, two groups of rats were given lovastatin capsule 20 mg / kg and 0.5 CMC-Na solution by single dose. Plasma samples were collected at different time points before and after administration, and the plasma concentration was determined by liquid chromatography-mass spectrometry (LC-MS). Results Long-term administration of glycyrrhiza resulted in a significant increase in plasma exposure of lovastatin metabolite lovastatin in hyperlipidemic rats.
