甘草對高脂血癥大鼠模型洛伐他汀藥代動力學的影響

【摘要】:目的研究高脂血癥大鼠模型中甘草對洛伐他汀藥代動力學過程的影響。方法將18只SD大鼠隨機分為對照組(n=6)和實驗組(n=12)。實驗組采用高脂飼料喂養(yǎng)大鼠,構建高脂血癥大鼠模型,隨機分為洛伐他汀單獨給藥組(n=6)和洛伐他汀與甘草聯(lián)合給藥組(n=6)。聯(lián)合給藥組給予甘草顆粒,單獨給藥組給予相應體積的生理鹽水,連續(xù)7 d后,2組大鼠單劑量灌胃給予洛伐他汀膠囊(20 mg/kg,0.5%CMC-Na溶液)。分別于給藥前和給藥后不同時間點采集血漿樣品,采用液相色譜-質譜聯(lián)用方法測定血藥濃度,計算藥代動力學參數,并進行組間比較。結果長期給予甘草導致洛伐他汀代謝產物洛伐他汀酸在高脂血癥大鼠中血漿暴露水平顯著升高,聯(lián)合給藥組平均C_(max)較單獨給藥組升高約80%(P0.05),而AUC0-t和AUC0-∞分別升高115%和109%(P=0.005和P=0.027)。聯(lián)合給藥組與單獨給藥組比較,洛伐他汀的C_(max)和AUC增加,但無統(tǒng)計學差異(P0.05)。結論甘草可抑制洛伐他汀在高脂血癥大鼠體內的代謝,增加其體內暴露水平。
[Abstract]:Objective to study the effect of licorice on the pharmacokinetic process of lovastatin in hyperlipidemic rats. Methods Eighteen SD rats were randomly divided into two groups: control group (n = 18) and experimental group (n = 12). The experimental group was fed with hyperlipidemic diet to establish a hyperlipidemic rat model. The rats were randomly divided into two groups: lovastatin group (n = 6) and lovastatin / licorice group (n = 6). The combination group was given licorice granules, and the control group was given a corresponding volume of normal saline. After 7 days, two groups of rats were given lovastatin capsule 20 mg / kg and 0.5 CMC-Na solution by single dose. Plasma samples were collected at different time points before and after administration, and the plasma concentration was determined by liquid chromatography-mass spectrometry (LC-MS). Results Long-term administration of glycyrrhiza resulted in a significant increase in plasma exposure of lovastatin metabolite lovastatin in hyperlipidemic rats.
