【摘要】：目的:研究E1A激活基因阻遏子(Cellular repressor of E1A-stimulated genes,CREG)對小鼠腹腔巨噬細胞溶酶體發(fā)生及溶酶體組織蛋白酶表達的影響。方法:應用loss-of-function和gain-of-function模型,Lysotracker Red染色和透射電鏡觀察CREG對小鼠腹腔巨噬細胞溶酶體發(fā)生的影響,并通過細胞免疫熒光染色和Western blot方法,觀察CREG對小鼠腹腔巨噬細胞溶酶體組織蛋白酶表達的影響。結果:Lysotracker Red染色和透射電鏡證實CREG可促進小鼠腹腔巨噬細胞溶酶體發(fā)生;細胞免疫熒光染色和Western blot方法證實CREG可促進小鼠腹腔巨噬細胞溶酶體組織蛋白酶cathepsin B及cathepsin S表達。結論:CREG可促進小鼠腹腔巨噬細胞溶酶體發(fā)生及組織蛋白酶cathepsin B及cathepsin S表達。
[Abstract]:Objective: to study the effects of Cellular repressor of E1A-stimulated genesutrid (CREGG) on lysosome formation and lysosomal cathepsin expression in murine peritoneal macrophages. Methods: the expression of lysosomal cathepsin in murine peritoneal macrophages was observed by loss-of-function and gain-of-function Red staining and transmission electron microscopy (TEM). Effects of lysosomes on peritoneal macrophages in mice, The effect of CREG on the expression of lysosomal cathepsin in murine peritoneal macrophages was observed by cell immunofluorescence staining and Western blot staining. The expression of lysosomal cathepsin B and cathepsin S in murine peritoneal macrophages was enhanced by CREG by cell immunofluorescence staining and Western blot method. Conclusion the expression of cathepsin B and cathepsin S in murine peritoneal macrophages can be enhanced by the expression of cathepsin B and cathepsin S in murine peritoneal macrophages.
