紫蘇干預(yù)COPD系統(tǒng)性炎癥的大鼠模型研究

【摘要】:為揭示導(dǎo)入紫蘇基因的新型煙草的生物學(xué)效應(yīng),將54只清潔級雄性SD大鼠隨機分為健康對照組(CK)、普通煙草組(GT)和新型煙草組(NT),采用煙霧熏吸法構(gòu)建COPD模型。對血清進(jìn)行ELISA分析顯示,香煙煙霧染毒56 d能誘發(fā)COPD大鼠的系統(tǒng)性炎癥反應(yīng);但與GT組相比,NT組的CRP、IL-8以及TNF-α水平顯著降低(P0.05)。取肝和脾組織進(jìn)行IHC檢測發(fā)現(xiàn),COPD大鼠的NF-κB p65表達(dá)水平明顯升高,且GT組與NT組脾組織間的差異顯著(P0.05)。結(jié)果表明,導(dǎo)入紫蘇基因的新型煙草能夠抑制大鼠吸煙相關(guān)性COPD的全身性炎癥反應(yīng),干預(yù)肺外組織NF-κB的過度活化,延緩疾病的進(jìn)程。
[Abstract]:In order to reveal the biological effects of new tobacco introduced perilla gene, Fifty-four clean male Sprague-Dawley rats were randomly divided into healthy control group, common tobacco group and new tobacco group. The COPD model was established by smoke fumigation. The ELISA analysis of serum was performed. The systemic inflammatory response of COPD rats was induced by cigarette smoke exposure for 56 days, but the levels of IL-8 and TNF- 偽 in NT group were significantly lower than those in GT group. The expression of NF- 魏 B p65 in liver and spleen tissues was significantly higher than that in GT group, and the expression level of NF- 魏 B p65 in liver and spleen tissues was significantly increased after exposure to cigarette smoke for 56 days. The difference between GT group and NT group was significant (P 0.05). The results showed that the new tobacco introduced perilla gene could inhibit the systemic inflammation of smoking-related COPD in rats, interfere with the excessive activation of NF- 魏 B in extrapulmonary tissue, and delay the progression of the disease.
