【摘要】：目的探討唑來(lái)膦酸對(duì)骨質(zhì)疏松模型大鼠骨密度及骨代謝的影響。方法將48只雌性SD大鼠按照隨機(jī)數(shù)字法隨機(jī)分為空白對(duì)照組、模型對(duì)照組、低劑量組(5 mg/kg)及高劑量組(10 mg/kg),每組12只。治療結(jié)束12 w后,測(cè)定大鼠股骨骨密度、股骨抗彎力、子宮指數(shù)及雙腎指數(shù),檢測(cè)血清骨鈣素、1,25-二羥基維生素D3〔1,25(OH)2D3〕、血鈣、血磷水平。結(jié)果與空白對(duì)照組比較,模型對(duì)照組及低劑量組大鼠骨密度、抗彎力、血清骨鈣素、1,25(OH)2D3、血鈣、子宮指數(shù)及雙腎指數(shù)明顯偏低(P0.05),而高劑量與空白對(duì)照組比較無(wú)統(tǒng)計(jì)學(xué)差異(P0.05);與模型對(duì)照組比較,低劑量組和高劑量組大鼠骨密度、抗彎力、血清骨鈣素、1,25(OH)2D3、血鈣、子宮指數(shù)及雙腎指數(shù)明顯偏高(P0.05);與低劑量組比較,高劑量組明顯偏高(P0.05)。結(jié)論唑來(lái)膦酸10 mg/kg劑量能明顯改善骨質(zhì)疏松模型大鼠的骨密度及骨代謝水平,升高子宮及雙腎指數(shù),對(duì)骨質(zhì)疏松模型大鼠具有較好的治療效果。
[Abstract]:Objective to investigate the effects of zoledronic acid on bone mineral density and bone metabolism in osteoporosis model rats. Methods 48 female SD rats were randomly divided into control group and model control group. Low dose group (5 mg / kg) and high dose group (10 mg / kg), 12 rats in each group. After 12 weeks of treatment, bone mineral density (BMD), bending strength of femur, uterine index and double kidney index were measured, serum osteocalcin 125-dihydroxyvitamin D ~ (3 +) ~ (25) OH _ (2) D _ (3), serum calcium, and serum calcium were measured. Results compared with the blank control group, the bone mineral density (BMD), bending resistance, serum osteocalcin (OHH) 2D3 and serum calcium in the model control group and low dose group were higher than those in the control group. The uterine index and the double kidney index were significantly lower than that of the control group, but there was no significant difference between the high dose group and the blank control group (P 0.05), compared with the model control group, the bone mineral density (BMD), the bending resistance, the serum osteocalcin (BGP) of the rats in the low dose group and the high dose group, and the serum calcium content were higher than those in the control group. Compared with the low dose group, the uterine index and the double kidney index were significantly higher in the high dose group than in the low dose group. Conclusion the 10 mg/kg dose of zoledronic acid can improve the bone mineral density and bone metabolism, increase the index of uterus and kidney, and increase the index of uterus and kidney in the model rats with osteoporosis. It has good therapeutic effect on osteoporosis model rats.
