Lnc-CLRN3與腸道天然免疫的相關(guān)性及其功能研究

【摘要】:目的研究一個全新的長鏈非編碼RNA Lnc-CLRN3在腸道天然免疫中的作用。方法應(yīng)用脂多糖(lipopolysaccharide,LPS)刺激人結(jié)腸癌細胞SW480,然后收集細胞提取RNA并通過RT-qPCR篩選表達變化的長鏈非編碼RNA,進一步通過基因過表達、敲除和RNA免疫共沉淀等技術(shù)手段對該基因的功能進行深入分析。結(jié)果篩選出1個在SW480細胞中高表達且LPS刺激基因表達上調(diào)的長鏈非編碼RNA Lnc-CLRN3,該基因主要分布于細胞核內(nèi),其轉(zhuǎn)錄受到MAPK通路的調(diào)控,進一步研究表明該基因通過結(jié)合SFPQ蛋白進而影響下游IL12P40等免疫因子的表達。結(jié)論長鏈非編碼RNA Lnc-CLRN3參與了腸道細胞的天然免疫調(diào)控。
[Abstract]:Objective to study the role of a novel long chain noncoding RNA Lnc-CLRN3 in intestinal innate immunity. Methods Lipopolysaccharide lipopolysaccharide (LPS) was used to stimulate human colon cancer cell line SW480, and then the cells were collected to extract RNA and to screen the long chain noncoding for the change of expression by RT-qPCR. RNAs, further overexpression of genes, The function of the gene was analyzed by knockout and RNA immunoprecipitation. Results A long chain non-coding RNA Lnc-CLRN3, which was highly expressed in SW480 cells and up-regulated by LPS stimulating gene, was selected, and the gene was mainly distributed in the nucleus. Its transcription is regulated by MAPK pathway. Further studies indicate that the gene affects the expression of downstream IL12P40 by binding SFPQ protein. Conclusion long chain non-coding RNA Lnc-CLRN3 is involved in the innate immune regulation of intestinal cells.
