【摘要】：目的研究復(fù)方大承氣湯灌服和芒硝、冰片、吳茱萸適量混合后于臍眼外敷預(yù)防大鼠模型術(shù)后腸粘連的作用及機(jī)制。方法雄性SD大鼠120只,隨機(jī)分為對照組、模型組、醫(yī)用幾丁糖組、生長抑素組、中草藥一組和中草藥二組。除正常對照組外,其余各組大鼠均采用Ellis法制備腸粘連模型,造模后對照組和模型組正常喂養(yǎng),其余各組分別給予相應(yīng)藥物:醫(yī)用幾丁糖組在造模關(guān)腹時局部涂抹幾丁糖;生長抑素組給予皮下注射醋酸奧曲肽0.1μg/g,1次/12 h;中草藥一組給予復(fù)方大承氣湯煎液0.05 mL/g灌服,2次/d;中草藥二組在一組用藥的基礎(chǔ)上,用芒硝、冰片、吳茱萸適量混合后外敷于臍眼。各組于造模術(shù)后第8天下腔靜脈取血后處死,ELISA法測定血清腫瘤壞死因子-α(TNF-α)含量,肉眼觀察腸粘連的程度并分級,采用免疫組化法測定腹膜粘連組織轉(zhuǎn)化生長因子-β_1(TGF-β_1)表達(dá)。結(jié)果與模型組對比,醫(yī)用幾丁糖組、生長抑素組、中草藥一組和中草藥二組大鼠血清中TNF-α含量依次降低,局部粘連程度依次減輕,粘連組織中TGF-β_1表達(dá)依次減少,中草藥一組和二組與模型組、醫(yī)用幾丁糖組、生長抑素組相比差異有統(tǒng)計學(xué)意義(P0.05),中草藥二組和中草藥一組之間相比差異有統(tǒng)計學(xué)意義(P0.05),醫(yī)用幾丁糖組和生長抑素組間差異無統(tǒng)計學(xué)意義(P0.05)。結(jié)論聯(lián)合應(yīng)用復(fù)方大承氣湯灌服和芒硝、冰片、吳茱萸適量混合后于臍眼外敷能顯著抑制纖維結(jié)締組織的增生,減輕造模大鼠模型局部粘連程度。
[Abstract]:Objective to study the effect and mechanism of compound Dachengqi decoction (Dachengqi decoction) on preventing intestinal adhesion after umbilical eye application. Methods 120 male Sprague-Dawley rats were randomly divided into control group and model group. Except for the normal control group, the other groups of rats made intestinal adhesion model by Ellis method, the control group and the model group were fed normally after the establishment of the model. The other groups were given corresponding drugs: chitosan was applied locally in the medical chitosan group when the model was closed; The somatostatin group was subcutaneously injected with octreotide acetate 0.1 渭 g / g for 12 h; the Chinese herbal medicine group 1 was given the compound Dachengqi decoction 0.05 mL/g decoction for 2 times per day; the second group was treated with mirabilite and borneol on the basis of one group. Evodia rutaecarpa was mixed with appropriate amount and applied to the umbilical eye. The serum levels of TNF- 偽 TNF- 偽 were determined by Elisa after the 8th operation of the model, and the degree and grade of intestinal adhesion were observed with naked eyes. The expression of TGF- 尾 in peritoneal adhesion tissue was determined by immunohistochemical method. Results compared with the model group, the serum TNF- 偽 levels in the medical chitosan group, somatostatin group, Chinese herbal medicine group and Chinese herbal medicine group were decreased in turn. The degree of local adhesion was reduced in turn, the expression of TGF- 尾 1 in adhesive tissue was decreased in turn, the Chinese herbal medicine group 1 and 2 and the model group, the medical chitosan group, There were significant differences in somatostatin group (P 0.05), Chinese herbal medicine group (2 groups) and Chinese herbal medicine group (1 group). There was no significant difference between medical chitosan group and somatostatin group (P 0.05). Dachengqi decoction and mirabilite, The mixture of borneol and rutaecarpa could significantly inhibit the proliferation of fibrous connective tissue and reduce the degree of local adhesion of rat model.
