猬迭宮絳蟲裂頭蚴感染小鼠皮下肌肉組織β1轉化生長因子含量變化的觀察

【摘要】:目的探討β1轉化生長因子(TGF-β1)在猬迭宮絳蟲(Spirometra erinacei)裂頭蚴感染小鼠皮下肌肉組中的表達情況及意義。方法對采自野生王錦蛇(Elaphe carinata)的裂頭蚴進行形態(tài)學觀察和PCR檢測。昆明小鼠80只,用數(shù)字隨機表法挑選40只,雌雄各半作為實驗組,剩余40只為對照組。用蛇源裂頭蚴經口喂飼實驗組小鼠,5條/鼠。于喂飼后第7、14、28、56天各隨機剖殺10只,分別收集含有裂頭蚴寄生的皮下肌肉組織,用中性甲醛固定,制作石蠟切片。HE染色后,觀察裂頭蚴感染病灶周圍的病理變化和纖維化程度;免疫組化檢測皮下肌肉中TGF-β1的含量變化。對照組不喂飼裂頭蚴,檢測時間及方法同實驗組。結果PCR擴增獲得約400 bp的目的條帶。測序結果顯示,該裂頭蚴COX1基因與Gen Bank中的猬迭宮絳蟲序列一致性為99.12%。HE染色檢查,皮下肌肉中的裂頭蚴被炎性囊壁所包裹,蟲體與囊壁之間形成穴腔,穴腔有時出現(xiàn)少許的漿液或血液。囊壁最里面的區(qū)域有薄層的纖維蛋白、壞死的碎片,壁中早期以中性粒細胞浸潤為主,也有嗜酸粒細胞、巨噬細胞、漿細胞等,隨著病程的進展,炎性囊壁逐漸擴大,壁中以慢性炎性細胞浸潤為主,囊壁周圍是宿主的組織細胞,纖維結締組織增生明顯,并有不同程度的纖維化。免疫組化檢查結果顯示,TGF-β1的主要表達部位為裂頭蚴周圍的炎癥反應帶和纖維結締組織增生部位。TGF-β1相對表達量在感染后逐漸增高,于感染第28天達峰值(0.654 5±0.045 5),第56天時明顯下降。在感染后第7~56天,實驗組的TGF-β1水平為(0.502 6±0.008 2)~(0.346 8±0.030 4)與同期對照組的(0.270 0±0.001 6)~(0.274 0±0.005 1)比較,差異均有統(tǒng)計學意義(P0.05)。結論小鼠感染猬迭宮絳蟲裂頭蚴早、中期,TGF-β1表達水平均提高,其免疫抑制作用不利于清除和控制皮下肌肉組織中的裂頭蚴。
[Abstract]:Objective to investigate the transforming growth factor beta 1 (TGF- beta 1) in Spirometra erinaceieuropaei (Spirometra erinacei) crack expression and the significance of subcutaneous muscle group in plerocercoid infection. Methods collected from wild Wang Jinshe (Elaphe carinata) the sparganum morphological observation and PCR detection. 80 Kunming mice, using digital randomly selected 40 rats, male and female as the experimental group, the remaining 40 rats as the control group. With the snake source sparganum oral feeding in the experimental group, 5 rats were fed to /. 7,14,28,56 days after the random killing of 10, were collected from muscle tissue containing sparganum parasitic skin next, fixed by formalin, paraffin sections after.HE staining, observe pathological changes and fibrosis degree of fissure lesions around plerocercoid infection; immunohistochemical changes of the content of TGF- beta 1 detection of subcutaneous muscle. The control group fed with sparganum, detection time and the results of PCR method with the experimental group. About 400 BP amplified purpose bands. The sequencing results showed that the sparganum COX1 gene and Gen Bank of Spirometra erinaceieuropaei sequence consistency for 99.12%.HE staining, subcutaneous sparganosis in muscle surrounded by inflammatory cystic wall, cavity formed between the body and the cystic wall, a little serous or blood cavity. The cyst wall has the innermost region of fibrin layer, necrotic debris, wall early by neutrophils, also have eosinophils, macrophages, plasma cells, with the progression of inflammatory cystic wall in chronic gradually expanded. Inflammatory cell infiltration, cystic wall surrounding host tissue, connective tissue hyperplasia, and there are different degrees of fibrosis. Immunohistochemical examination showed that TGF- mainly expressed in parts of the beta 1 inflammation sparganum surrounding connective tissue hyperplasia with fiber and parts .TGF- beta 1 expression increased gradually after infection, infection in twenty-eighth Tianda peak (0.6545 + 0.0455), the fifty-sixth day decreased significantly. At 7~56 days after infection, the experimental group TGF- beta 1 level (0.5026 + 0.0082) ~ (0.3468 + 0.0304) compared with the control group (0.2700 + 0.0016) ~ (0.2740 + 0.0051), the differences were statistically significant (P0.05). Conclusion the mice infected with sparganosis Spirometra erinaceieuropaei early, mid, TGF- beta 1 expression levels were increased, the immunosuppressive effect is not conducive to the removal and control of subcutaneous sparganosis in muscle tissue.
