【摘要】：目的探討建立剛地弓形蟲穩(wěn)定垂直傳播小鼠模型的方法。方法將42只孕8dBALB/c小鼠隨機分為(A-F組),正常對照A組(孕鼠經(jīng)腹腔注射無菌PBS 0.2ml/只)和感染組(B~F)(每只小鼠經(jīng)腹腔接種不同劑量的弓形蟲速殖子10、20、40、80個/0.2ml)。觀察并記錄孕鼠的體重改變、行為反應、流產(chǎn)率與活胎率;孕18d時剖腹,取出胎鼠進行形態(tài)學觀察。結果感染組小鼠出現(xiàn)倦怠、豎毛、弓背等異常反應,且隨速殖子感染數(shù)量的增加異常反應程度逐漸嚴重;F組孕鼠死亡2只。感染組較對照組體重增加不明顯,F組體重減輕;孕18d時D~F組體重與對照組比較差異有統(tǒng)計學意義(P0.05)。感染組出現(xiàn)流產(chǎn)、死胎等,F組存活孕鼠腹中無胎鼠。D~F組流產(chǎn)率與對照組相比差異有統(tǒng)計學意義(P0.05);B~F組活胎率與對照組比較差異有統(tǒng)計學意義(P0.05)。E~F組胎鼠體重、體長、尾長與對照組比較差異有統(tǒng)計學意義(P0.05)。結論成功建立剛地弓形蟲垂直傳播小鼠模型,BALB/c小鼠孕8d腹腔注射接種RH株弓形蟲速殖子40個/(0.2ml·只)為適宜感染劑量。
[Abstract]:Objective to establish a stable vertical transmission model of Toxoplasma gondii (Toxoplasma gondii). Normal control group A (pregnant rats injected aseptic PBS 0.2ml / mouse) and infected group (each mouse intraperitoneally inoculated with Toxoplasma gondii Toxoplasma Toxoplasma Toxoplasma Tachyzoites 1020M40). 80 / 0.2ml. the body weight change, behavioral reaction, abortion rate and live birth rate of pregnant rats were observed and recorded. On the 18th day of gestation, the fetal mice were taken out for morphological observation. Results in the infected group, abnormal reactions such as burnout, bristles and back of the bow were observed, and the abnormal reaction degree was gradually serious with the increase of tachyzoite infection. Two pregnant rats died in group F. the weight loss of infection group was not significant compared with that of control group. At the 18th day of gestation, there was a significant difference in body weight between DU F group and control group (P 0.05). Abortion and stillbirth occurred in infection group. The abortion rate of group F was significantly higher than that of control group (P 0.05). Compared with the control group, there was a significant difference in the rate of live fetus in group B and F (P 0.05). The body length and body weight of fetal rats in group E were significantly higher than those in group B (P 0.05). Conclusion the vertical transmission model of Toxoplasma gondii was established successfully. BALB/c mice were injected intraperitoneally with RH strain Toxoplasma gondii Toxoplasma gondii Toxoplasma gondii Toxoplasma gondii 40 / 0. 2ml 路mouse) on the 8th day of gestation.
