去卵巢結(jié)合蔗糖鐵誘導(dǎo)家兔鐵蓄積狀態(tài)骨質(zhì)疏松模型的建立與評(píng)價(jià)

【摘要】:目的通過去卵巢結(jié)合蔗糖鐵誘導(dǎo)家兔鐵蓄積狀態(tài)骨質(zhì)疏松模型,以期為有關(guān)鐵蓄積狀態(tài)骨質(zhì)疏松的實(shí)驗(yàn)研究提供動(dòng)物模型基礎(chǔ)。方法將30只5月齡的新西蘭雌性家兔隨機(jī)分為對(duì)照組、去勢組和鐵蓄積組;每組分別于術(shù)前1天檢測血清1型前膠原氨基末端肽(P1NP),1型膠原羥基末端肽(CTX)及血清鐵蛋白水平,并行腰椎骨密度測量;去勢組和鐵蓄積組手術(shù)切除雙側(cè)卵巢,對(duì)照組僅切開卵巢周圍部分脂肪;1周后鐵蓄積組耳緣靜脈注射蔗糖鐵,劑量為15 mg/kg,每周1次,共12周;對(duì)照組和去勢組以同樣方式和頻次給予等量生理鹽水注射;所有組分別于術(shù)后4、8、12、16、20、24周檢測血清P1NP,CTX及血清鐵蛋白水平,并行腰椎骨密度測量。結(jié)果 (1)所有家兔注射蔗糖鐵后均未出現(xiàn)昏迷、抽搐、精神萎靡、呼吸急促等不良反應(yīng)或致死的情況;(2)鐵蓄積組血清鐵蛋白水平在造模后8、12、16、20及24周均明顯高于對(duì)照組(P0.05);(3)鐵蓄積組與去勢組血清P1NP,CTX水平在造模后12周、16周、20周及24周均明顯高于對(duì)照組(P0.05);鐵蓄積組血清P1NP,CTX水平在造模后20周及24周均明顯高于去勢組(P0.05);(4)鐵蓄積組與去勢組腰椎骨密度丟失百分率均在造模后第24周超過25%(33.72%、25.92%)。結(jié)論去卵巢結(jié)合蔗糖鐵可成功建立家兔鐵蓄積狀態(tài)骨質(zhì)疏松模型。
[Abstract]:Objective to induce the model of iron accumulation in rabbits by ovariectomized with sucrose iron. Methods Thirty 5-month-old New Zealand female rabbits were randomly divided into control group, castrated group and iron accumulation group. Serum procollagen type 1 amino terminal peptide (P1NPN) and serum ferritin (CTX) were measured 1 day before operation in each group. The bone mineral density of lumbar vertebrae was measured. The ovariectomized group and the iron accumulation group had bilateral ovariectomies, while the control group only cut some fat around the ovary. One week later, sucrose iron was injected intravenously into the ear margin of the iron accumulation group at a dose of 15 mg / kg once a week for 12 weeks. The control group and ovariectomized group were injected with the same amount of saline in the same way and frequency. The levels of serum P1NPCTX and serum ferritin were measured in all groups at 24 weeks after operation. Results all rabbits had no adverse reaction or death after injection of iron sucrose, such as coma, convulsion, mental retardation, shortness of breath, etc. (2) the level of serum ferritin in the iron accumulation group was significantly higher than that in the control group at 20 and 24 weeks after the establishment of the model. (3) the levels of serum P1NPN CTX in the iron accumulation group and castration group were significantly higher than those in the control group at the 12th week, the 16th week, the 20th week and the 24th week, respectively. The level of serum P1NPN CTX in the iron accumulation group was significantly higher than that in the castrated group at 20 and 24 weeks after the establishment of the model. 4) the percentage of bone mineral density loss of lumbar vertebrae in the iron accumulation group and castration group was more than 33.72% at the 24th week after the establishment of the model. Conclusion Ovariectomized with sucrose iron can successfully establish the model of iron accumulation in rabbits.
