玉米須提取物對(duì)糖尿病足小鼠模型血糖、血脂及血液流變學(xué)的影響

【摘要】:目的:探討玉米須提取物對(duì)鏈脲佐菌素(STZ)誘導(dǎo)的糖尿病足模型血糖、血脂及血液流變學(xué)的影響。方法:昆明種小鼠150只,適應(yīng)性飼養(yǎng)1周,隨機(jī)將150只小鼠分成5組,30只/組,分別為空白組、模型對(duì)照組、玉米須提取液高劑量組、中劑量組、低劑量組?瞻捉M喂以標(biāo)準(zhǔn)飼料,除空白組外其余組喂以高脂高糖飲食至實(shí)驗(yàn)結(jié)束。11周后除空白組外均造模,造模前禁食12 h,自由飲水,按每天40mg/kg劑量,將STZ用濃度為0.1 mmol/L檸檬酸緩沖液(p H4.2)溶解,給小鼠做腹腔注射1次/d,連續(xù)5天。除空白組和模型對(duì)照組外,其余3組玉米須提取物按照400、200、100 mg/kg,配成濃度為20、10、5g/L,灌胃體積均為0.2 m L/10 g灌胃給藥。給藥第30天,給藥后1 h,禁食12 h,小鼠尾部取血,觀察小鼠用藥前后的血糖、血脂及血液流變學(xué)的指標(biāo)變化。結(jié)果:給藥后玉米須提取物高、中、低劑量組與模型對(duì)照組相比較均具有降低糖尿病足小鼠模型血糖的作用P0.01,且呈現(xiàn)劑量依賴性,組間有差異P0.05,高劑量組要明顯優(yōu)于低劑量組的降糖效果;給藥后高、中、低劑量組與模型對(duì)照組相比較均具有改善血脂、血液流變學(xué)的作用P0.05,且呈現(xiàn)劑量依賴性,組間有差異P0.05,高劑量組要明顯優(yōu)于低劑量組改善血脂、血液流變學(xué)的作用。結(jié)論:玉米須提取物能夠有效改善小鼠糖尿病足模型血糖、血脂、血液流變學(xué)的作用,且改善的效果隨著劑量濃度的增高而增加。
[Abstract]:Objective: to investigate the effects of Corn whisker extract on blood glucose, blood lipid and hemorheology of diabetic foot model induced by streptozotocin (STZ). Methods: 150 Kunming mice were fed adaptively for one week. 150 mice were randomly divided into 5 groups (n = 30): blank group, model control group, high dose group, middle dose group and low dose group. The rest groups except the blank group were fed with high fat and high sugar diet until the end of the experiment. After 11 weeks, all the models were made except the blank group, fasting for 12 hours before making the model, drinking freely, at the dose of 40mg / kg per day. STZ was dissolved with 0.1 mmol/L citric acid buffer solution (pH4.2). Mice were injected intraperitoneally for 5 days, except blank group and model control group. The other three groups were treated with 400,200,100 mg / kg corn hair extract with a concentration of 20 ~ 10g / L ~ (-1) ~ (-1) ~ (-1) ~ (-1) 路kg ~ (-1). The volume of gavage was 0.2 mL / 10 g by gavage. On the 30th day, 1 hour after administration, 12 hours after fasting, blood was taken from tail of mice and blood glucose was observed before and after administration. Results: compared with the model control group, the serum lipids and hemorheology of the low and middle dose groups had the effect of reducing the blood glucose of diabetic foot mice (P0.01). There was a dose-dependent difference between the two groups (P0.05). The effect of high dose group was better than that of low dose group (P 0.05). Compared with the model control group, the high, medium and low dose groups had the effect of improving blood lipids and hemorheology (P0.05) after administration, and showed dose-dependent, the difference between the groups (P0.05). The effect of high dose group was better than that of low dose group in improving blood lipids and hemorheology. Conclusion: Corn whisker extract can effectively improve blood sugar, blood lipid and hemorheology of mice diabetic foot model. And the effect of improvement increased with the increase of dose concentration.
