【摘要】:目的探討發(fā)光二極管(Light-emitting Liode,LED)發(fā)射的波長620nm的低能量紅光對SD大鼠骨髓間充質(zhì)干細胞(Bone Mesenchymal Stem Cells,BMSCs)增殖的影響。 方法采用密度梯度離心法分離SD大鼠BMSCs,再經(jīng)貼壁法篩選,取生長良好,呈對數(shù)生長期的第3代BMSCs,波長620nm的LEDs置于細胞上方2cm處,測得功率密度為6.67mW/cm2。根據(jù)不同照射時間所對應的能量密度,將BMSCs分別設(shè)置0、0.5、1和2J/cm2組,其中0J/cm2組為對照組,其他組為實驗組,紅光每天照射1次。采用細胞計數(shù)試劑盒-8(CCK-8)檢測各組細胞紅光照射前3天的細胞增殖活性,EdU(5-Ethynyl-2-deoxyundine)細胞增殖檢測試劑盒和流式細胞儀分別檢測各組細胞紅光照射第3天后,DNA復制活性和細胞周期變化。 結(jié)果BMSCs經(jīng)620nm低能量紅光照射第3天后,實驗組細胞與對照組的相比,增殖活性和DNA復制活性顯著增強,(S、G2-M)期細胞數(shù)量顯著增多,差異具有統(tǒng)計學意義(p0.05),其中0.5J/cm2組效果最顯著。 結(jié)論LED發(fā)射的620nm低能量紅光照射SD大鼠BMSCs第3天后,可有效促進其體外增殖。
[Abstract]:Objective to study the Light-fitting Liode of light emitting diodes (LEDs). The bone Mesenchymal Stem Cells of bone marrow mesenchymal stem cells (BMSCs) of SD rats was induced by low-energy red light at 620nm. Effects of BMSCs) on proliferation. Methods BMSCs were isolated from SD rats by density gradient centrifugation. The third generation BMSCs with good growth and logarithmic growth was selected by adherent method. The wavelength of 620nm LEDs was placed at 2 cm above the cell, and the power density was 6.67 MW / cm ~ (2). According to the energy density corresponding to different irradiation time, the BMSCs was set at 0. The 0. 5 J / cm2 and 2 J / cm2 groups were the control group and the other groups were the experimental group. The cell proliferation activity of each group was detected 3 days before red light irradiation by cell counting kit -8 CCK-8. The proliferation test kit and flow cytometry were used to detect the red light of each group after 3 days of irradiation. DNA replication activity and cell cycle changes. Results after BMSCs was irradiated with 620nm low energy red light for 3 days, the proliferative activity and DNA replication activity of the experimental group were significantly increased compared with those of the control group. The number of G2-Mphase cells increased significantly, and the difference was statistically significant (P 0.05). The effect of 0.5 J / cm 2 group was the most significant. Conclusion LED emission of 620nm low energy red light irradiation on the third day of BMSCs in SD rats can effectively promote the proliferation of SD rats in vitro.
