高脂飲食對(duì)NAFLD模型大鼠腸道菌群及血清LPS水平的影響

【摘要】:目的觀(guān)察高脂飲食誘導(dǎo)的非酒精性脂肪肝病(NAFLD)模型大鼠腸道菌群及血清內(nèi)毒素(LPS)水平的變化,探討腸道菌群改變及LPS血癥在NAFLD發(fā)病過(guò)程中的作用。方法雄性SD大鼠12只隨機(jī)分為對(duì)照組和高脂組,對(duì)照組以普通飲食飼養(yǎng),高脂組給予高脂飼料喂養(yǎng),于實(shí)驗(yàn)第16周末處死大鼠,采用全自動(dòng)生化分析儀測(cè)定血清生化指標(biāo),電鏡觀(guān)察兩組大鼠肝臟病理改變,ELISA法檢測(cè)血清LPS、白介素(IL)-1β、IL-18水平的變化,通過(guò)實(shí)時(shí)熒光定量PCR法檢測(cè)糞便大腸桿菌和乳酸桿菌的表達(dá)水平。結(jié)果高脂組大鼠血清谷丙轉(zhuǎn)氨酶、谷草轉(zhuǎn)氨酶、三酰甘油、總膽固醇、極低密度脂蛋白水平較對(duì)照組明顯升高,高密度脂蛋白水平明顯降低,差異均有統(tǒng)計(jì)學(xué)意義(P0.05);與對(duì)照組相比,高脂組大鼠血清LPS、IL-1β、IL-18水平明顯升高,差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。與對(duì)照組相比,高脂組大鼠的的大腸桿菌顯著升高,而乳酸桿菌明顯減少,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。血清LPS水平與大腸桿菌量存在相關(guān)性。結(jié)論高脂飲食誘導(dǎo)的NAFLD模型大鼠存在LPS血癥和腸道菌群的改變,不合理的飲食結(jié)構(gòu)及糞便菌群的改變?cè)贜AFLD的發(fā)病過(guò)程中起著重要的作用。
[Abstract]:Objective to observe the changes of intestinal flora and serum endotoxin level in NAFLD- induced non-alcoholic fatty liver disease rats induced by high fat diet. Methods Twelve male Sprague-Dawley rats were randomly divided into control group and hyperlipidemia group. The rats in the high-fat group were fed with high-fat diet. The rats were killed at the 16th week of the experiment. The serum biochemical indexes were measured by automatic biochemical analyzer and the pathological changes of liver were observed by electron microscope. The changes of serum LPSand IL-18 levels were detected by ELISA. Results Serum alanine aminotransferase, alanine aminotransferase, triacylglycerol and total cholesterol were detected by real-time fluorescence quantitative PCR. The level of very low density lipoprotein was significantly higher than that of the control group, and the level of high density lipoprotein was significantly lower than that of the control group (P 0.05). Compared with the control group, the serum levels of IL-1 尾 -IL-18 in the hyperlipidemia group were significantly higher than those in the control group (P 0.05). The Escherichia coli level in the high-fat group was significantly higher than that in the control group, while the number of Lactobacillus was significantly decreased. The difference was statistically significant (P 0.05). The level of serum LPS was correlated with the amount of Escherichia coli. Conclusion the NAFLD model rats induced by high fat diet have LPS and intestinal flora changes. Unreasonable diet and fecal microflora play an important role in the pathogenesis of NAFLD.
