骨髓間充質(zhì)干細(xì)胞移植對(duì)糖尿病模型大鼠腎臟的保護(hù)作用

【摘要】:背景:國(guó)內(nèi)外關(guān)于干細(xì)胞移植對(duì)糖尿病慢性并發(fā)癥的治療作用已有較多理論研究,但其對(duì)糖尿病腎病的治療作用尚無動(dòng)物實(shí)驗(yàn)證實(shí)。目的:觀察骨髓間充質(zhì)干細(xì)胞移植對(duì)糖尿病模型大鼠腎臟的保護(hù)作用。方法:將大鼠用高糖高脂飲食配方喂養(yǎng)4周后注射鏈脲佐菌素,建立2型糖尿病模型大鼠,造模成功的第2天開始經(jīng)尾靜脈注射骨髓間充質(zhì)干細(xì)胞單細(xì)胞懸液,并設(shè)對(duì)照組和糖尿病組大鼠作對(duì)照。細(xì)胞移植后第21天檢查大鼠尾靜脈血的血糖、三酰甘油和胰島素水平,并進(jìn)行腎臟和胰腺組織的病理學(xué)觀察。結(jié)果與結(jié)論:(1)生化指標(biāo):糖尿病組大鼠血糖、血清胰島素和三酰甘油水平均明顯高于對(duì)照組(P0.05)。干細(xì)胞移植組大鼠血糖和血清胰島素水平明顯低于糖尿病組(P0.05);(2)腎臟和胰腺組織的病理學(xué)變化:干細(xì)胞移植組大鼠腎小球系膜面積與腎小球體積均明顯小于糖尿病組(P0.05);(3)結(jié)果證實(shí):骨髓間充質(zhì)干細(xì)胞移植后可降低血糖及血清胰島素水平,修復(fù)受損的胰腺及腎臟損傷。
[Abstract]:Background: there have been many theoretical studies on the therapeutic effect of stem cell transplantation on diabetic chronic complications at home and abroad. But its therapeutic effect on diabetic nephropathy has not been confirmed by animal experiments. Objective: to observe the protective effect of bone marrow mesenchymal stem cell transplantation on kidney of diabetic model rats. Rats were fed with high sugar and high fat diet for 4 weeks, then streptozotocin was injected. Type 2 diabetic model rats were established and the single cell suspension of bone marrow mesenchymal stem cells was injected through caudal vein on the second day of successful modeling. The levels of blood glucose triglyceride and insulin in caudal vein blood of rats were examined 21 days after transplantation. The pathological changes of kidney and pancreas were observed. Results and conclusion the biochemical index: blood glucose in diabetic rats. The levels of serum insulin and triglyceride were significantly higher than those of the control group (P 0.05). The levels of blood glucose and serum insulin in the stem cell transplantation group were significantly lower than those in the diabetic group. (2) pathological changes of kidney and pancreas: the glomerular Mesangial area and glomerular volume in the stem cell transplantation group were significantly smaller than those in the diabetic group (P 0.05); The results showed that bone marrow mesenchymal stem cells (BMSCs) could reduce blood glucose and serum insulin level and repair the damaged pancreas and kidney injury after bone marrow mesenchymal stem cell transplantation.
