【摘要】:由腫瘤、外傷、炎癥等多種因素引起的頜骨缺損和吸收是口腔醫(yī)學(xué)領(lǐng)域比較常見的臨床難題。成骨細(xì)胞是參與骨形成的主要細(xì)胞,其前體細(xì)胞骨髓間充質(zhì)干細(xì)胞在適當(dāng)?shù)臈l件下能分化成多種細(xì)胞,如成軟骨細(xì)胞、成骨細(xì)胞、成脂肪細(xì)胞、成肌細(xì)胞等。通過對(duì)骨髓間充質(zhì)干細(xì)胞向成骨細(xì)胞的分化和功能調(diào)控的研究,不僅對(duì)正確認(rèn)識(shí)骨形成的機(jī)理具有重要的理論意義,而且對(duì)進(jìn)一步治療骨吸收相關(guān)疾病、促進(jìn)骨再生具有實(shí)際的指導(dǎo)意義。 骨形成是一個(gè)受多種細(xì)胞因子和生長(zhǎng)因子調(diào)控的過程,如何利用生長(zhǎng)因子更好的促進(jìn)骨組織愈合,是目前的研究熱點(diǎn)之一。研究表明,骨髓間充質(zhì)干細(xì)胞復(fù)合骨源性生長(zhǎng)因子能顯著促進(jìn)骨再生。BMP-2作為一種骨形成調(diào)控因子,在促進(jìn)骨折愈合和骨缺損修復(fù)過程中作用顯著。本實(shí)驗(yàn)擬通過轉(zhuǎn)基因方法將外源性BMP-2導(dǎo)入骨髓間充質(zhì)干細(xì)胞,使其高效、穩(wěn)定的表達(dá),以達(dá)到促進(jìn)骨形成的目的。 本研究通過建立體外骨髓間充質(zhì)干細(xì)胞培養(yǎng)方法,用所構(gòu)建的腺病毒載體攜帶BMP-2基因轉(zhuǎn)染體外培養(yǎng)的骨髓間充質(zhì)干細(xì)胞。通過MTT、堿性磷酸酶活性檢測(cè)和茜素紅染色等方法檢測(cè)BMP-2對(duì)骨髓間充質(zhì)干細(xì)胞增殖和成骨向分化的影響,結(jié)果表明BMP-2能促進(jìn)骨髓間充質(zhì)干細(xì)胞的增殖,并能促進(jìn)其向成骨細(xì)胞的分化。 因此,骨髓間充質(zhì)干細(xì)胞這一培養(yǎng)體系的建立為體外研究骨髓間充質(zhì)干細(xì)胞向成骨細(xì)胞分化和功能的調(diào)控提供了切實(shí)可行的方法;研究骨髓間充質(zhì)干細(xì)胞轉(zhuǎn)基因的可行性,為進(jìn)一步利用轉(zhuǎn)基因技術(shù)治療骨吸收性疾病、促進(jìn)骨形成奠定理論基礎(chǔ)。
[Abstract]:By the tumor, trauma, inflammation caused by a variety of factors such as the jaw defect and absorption is difficult in clinical dentistry more common. Osteoblasts are important cells in bone formation, the precursor cells of bone marrow mesenchymal stem cells can differentiate into multiple cell types in appropriate conditions, such as cartilage, bone cells, adipocytes, myoblasts. The bone marrow mesenchymal stem cells on the differentiation and function of regulation of bone cells, the mechanism not only has important theoretical significance on the formation of a correct understanding of bone, but also for the further treatment of bone resorption related diseases, promote bone regeneration has practical significance.
Bone formation by a variety of cytokines and growth factors regulating the process, how to make use of growth factors to better promote bone healing, is one of the hot research at present. The study shows that bone marrow mesenchymal stem cells combined with bone derived growth factor can significantly promote bone regeneration.BMP-2 as a regulatory factor in bone formation. To promote fracture healing and bone defect repair process significantly. The experiment of transgenic methods of exogenous BMP-2 bone marrow mesenchymal stem cells, to make it efficient and stable expression, in order to achieve the purpose of promoting bone formation.
In this study, through the establishment of in vitro culture of bone marrow mesenchymal stem cell by adenovirus vector constructed by carrying BMP-2 gene transfection in vitro cultured bone marrow mesenchymal stem cells. By MTT method, and detected by alizarin red staining and alkaline phosphatase activity detection of BMP-2 of bone marrow mesenchymal stem cells proliferation and osteogenic differentiation the results show that BMP-2 can promote the effect of bone marrow mesenchymal stem cell proliferation, and can promote the osteogenic differentiation.
Therefore, bone marrow mesenchymal stem cells in this culture system was established for the study of bone marrow mesenchymal stem cells and provides a feasible way to regulate osteoblast differentiation and function; study the feasibility of bone marrow mesenchymal stem cells genetically modified, for the further use of transgenic technology for the treatment of bone resorptive diseases, promote bone lay to form a theoretical basis.
