【摘要】：目的檢測雌雄小鼠肝組織蛋白質(zhì)組表達(dá)差異,探討肝病發(fā)生的性別差異機制。方法采用雙向熒光差異凝膠電泳(two dimension difference gel electrophoresis,2D-DIGE)和基質(zhì)輔助激光解吸電離飛行時間質(zhì)譜(matrix-assisted laser desorption/ionization time of flight mass spectrometry,MALDI-TOF-MS)技術(shù)檢測C57BL/6J雌雄小鼠肝組織的蛋白質(zhì)組差異表達(dá)譜,分離并鑒定差異表達(dá)蛋白,應(yīng)用Western blot進行驗證,并對差異表達(dá)蛋白進行生物信息學(xué)分析,包括蛋白功能注釋、分類分析、京都基因與基因組百科全書通路分析。結(jié)果經(jīng)雙向熒光差異凝膠電泳-圖像分析得到1767個蛋白點,其中差異倍數(shù)≥1.5(P0.05)的蛋白點325個。從中選擇78個差異蛋白點進行MALDI-TOF-MS鑒定,得到48種差異性表達(dá)的蛋白質(zhì)。其中,與雌鼠相比,雄鼠肝組織中高表達(dá)的蛋白有14種,低表達(dá)的蛋白有34種。選取6個差異蛋白點進行Western blot驗證,證明了2D-DIGE結(jié)果的可靠性。GO分析結(jié)果顯示,涉及雌雄鼠肝組織中的差異蛋白在細(xì)胞組分、分子功能、生物學(xué)過程中的分布廣泛。KEGG通路分析發(fā)現(xiàn)差異蛋白涉及6條信號通路。結(jié)論 C57BL/6J雌雄小鼠肝組織的差異性蛋白表達(dá)的檢測結(jié)果為研究性別差異性肝病發(fā)生的分子機制提供了基礎(chǔ)生物學(xué)信息和有益的線索。
[Abstract]:Objective to detect the expression difference between male and female mice liver tissue proteome, explore the mechanism of gender differences in liver disease. Methods using two-dimensional difference gel electrophoresis (two dimension difference gel electrophoresis, 2D-DIGE) and matrix assisted laser desorption ionization time-of-flight mass spectrometry (matrix-assisted laser desorption/ionization time of flight mass spectrometry, MALDI-TOF-MS) differential expression of proteome in detecting C57BL/6J male and female mouse liver tissue, the expression of protein isolate and identify the differences in application of Western blot to verify, and the differentially expressed protein by bioinformatics analysis, including protein functional annotation, classification analysis, analysis of Kyoto Encyclopedia of genes and genomes pathway. Results by fluorescence two-dimensional difference gel electrophoresis image analysis of 1767 protein spots, among them the difference is more than 1.5 times (P0.05) 325 protein spots. Choose from MALDI-TOF-MS identified 78 differential protein spots, 48 proteins were differentially expressed. Among them, compared with female mice, the high expression of liver tissue in male rats there are 14 kinds of proteins, low expression of protein 34. Selected 6 differentially expressed protein spots were Western blot validation,.GO reliability analysis results show that 2D-DIGE the results show that relates to differences in male and female rats in liver tissue protein in the cellular component, molecular function, biological process in the widely distributed.KEGG pathway analysis showed that the differential proteins involved in 6 pathways. Conclusion C57BL/6J male and female mouse liver tissue to detect differentially expressed proteins occurred for the study of gender differences in the molecular mechanism of liver disease the basic biological information and useful clues.
