【摘要】：目的從不同時(shí)間點(diǎn)比較幾種不同的應(yīng)激方法建立焦慮大鼠模型的生物學(xué)特點(diǎn),為尋找最合適的造模方法提供實(shí)驗(yàn)依據(jù)。方法 70只大鼠隨機(jī)分為正常組、空瓶應(yīng)激組、慢性情緒應(yīng)激(chronic emotional stress,CES)組、束縛應(yīng)激3 h組、束縛應(yīng)激6 h組,分別進(jìn)行造模,于實(shí)驗(yàn)第7 d、14 d、21 d分別采用高架十字迷宮和場景恐懼測(cè)試測(cè)定大鼠的焦慮樣行為,曠場測(cè)試考察大鼠的焦慮或抑郁樣行為,強(qiáng)迫游泳實(shí)驗(yàn)測(cè)定大鼠的抑郁樣行為,采用Elisa試劑盒檢測(cè)大鼠海馬組織中5-HT、DA含量。結(jié)果焦慮樣行為學(xué)測(cè)試結(jié)果表明,第14天起,空瓶應(yīng)激組、CES組、束縛應(yīng)激6 h組大鼠開始產(chǎn)生焦慮樣行為,此后焦慮樣行為愈發(fā)明顯,強(qiáng)迫游泳實(shí)驗(yàn)結(jié)果表明,束縛6 h組大鼠的不動(dòng)時(shí)間于第7 d起即顯著增加(P0.05)。同時(shí),與正常大鼠比較,空瓶應(yīng)激組和CES組大鼠海馬5-HT、DA含量于第14天開始顯著上升(P0.05或P0.01)。結(jié)論在上述幾種焦慮模型中,束縛應(yīng)激3 h焦慮樣行為不明顯;束縛應(yīng)激6 h可能由于應(yīng)激時(shí)間過長,表現(xiàn)出抑郁樣行為。空瓶應(yīng)激和CES能成功建立焦慮大鼠模型,且其焦慮行為表征有一定差異,可根據(jù)不同的實(shí)驗(yàn)?zāi)康倪x擇對(duì)應(yīng)的模型方法。
[Abstract]:The biological characteristics of stress from different time points to compare different anxiety to establish rat model, to find the most suitable model to provide experimental basis. Methods 70 rats were randomly divided into normal group, empty bottle stress group, chronic emotional stress (chronic emotional, stress, CES) group, 3 h restraint stress 6 h group, restraint stress group, respectively. The model was made on the seventh D, 14 d and 21 d respectively with anxiety like behavior and elevated plus maze test were measured at the scene of fear, open field tests in rats with anxiety or depression like behavior, depression like behavior in rats by forced swimming experiment using 5-HT, Elisa kit, tissue of rat hippocampus. The detection of DA content in anxiety like behavior test results show that, the fourteenth day, the empty bottle stress group, CES group, 6 h restraint stress rats began to produce anxiety like behavior, anxiety like behavior after the more obvious Forced swimming, the experimental results show that the immobility time bound 6 h rats in seventh D increased significantly (P0.05). At the same time, compared with the normal rats, the empty bottle stress group and CES in hippocampus of rats in 5-HT group, the content of DA on the fourteenth day increased significantly (P0.05 or P0.01). In conclusion the several kinds of anxiety model, restraint stress 3 h anxiety like behavior is not obvious; restraint stress may be due to stress 6 h long time, showed depression like behavior. Empty bottle stress and CES can successfully establish rat models of anxiety, and the anxiety behavior is different, according to different objectives selection model the corresponding.
