體外激活的小膠質(zhì)細(xì)胞對(duì)神經(jīng)元細(xì)胞的作用研究

【摘要】:目的1.建立和完善大鼠腦皮質(zhì)小膠質(zhì)細(xì)胞原代培養(yǎng)的實(shí)驗(yàn)方法和條件;2.探索原代培養(yǎng)的小膠質(zhì)細(xì)胞經(jīng)脂多糖(LPS)體外激活后對(duì)神經(jīng)元細(xì)胞的影響;3.通過丁基苯酞(NBP)對(duì)小膠質(zhì)細(xì)胞激活過程的影響,探討國家Ⅰ類新藥NBP可能的新的作用靶點(diǎn)。 方法原代培養(yǎng)大鼠腦皮質(zhì)小膠質(zhì)細(xì)胞和神經(jīng)元細(xì)胞;利用LPS刺激小膠質(zhì)細(xì)胞,建立原代培養(yǎng)的小膠質(zhì)細(xì)胞體外激活的細(xì)胞模型;提取LPS激活的小膠質(zhì)細(xì)胞的培養(yǎng)基上清液作用于神經(jīng)元細(xì)胞,通過四甲基偶氮唑鹽(MTT)檢測(cè)神經(jīng)元細(xì)胞的存活率:在加入LPS作用前用NBP處理小膠質(zhì)細(xì)胞,通過觀察小膠質(zhì)細(xì)胞的形態(tài),ELISA法定量檢測(cè)炎性因子IL-1β、TNF-α及IL-10的釋放,硝酸還原法檢測(cè)炎性介質(zhì)一氧化氮(NO),以及MTT檢測(cè)神經(jīng)元細(xì)胞經(jīng)其培養(yǎng)基上清液作用后的存活率,綜合評(píng)估NBP對(duì)小膠質(zhì)細(xì)胞激活的影響。 結(jié)果成功培養(yǎng)出數(shù)量多、純度高的大鼠腦皮質(zhì)小膠質(zhì)細(xì)胞:LPS激活的小膠質(zhì)細(xì)胞的培養(yǎng)基上清液可致神經(jīng)元細(xì)胞的存活率下降;NBP處理后的小膠質(zhì)細(xì)胞經(jīng)LPS作用后,仍呈現(xiàn)為激活形態(tài),其釋放的NO減少,IL-1β、TNF-α及IL-10的含量無明顯變化,經(jīng)其培養(yǎng)基上清液作用后的神經(jīng)元細(xì)胞存活率較對(duì)照組升高,且丁基苯酞的上述作用呈現(xiàn)一定的劑量依賴性。 結(jié)論激活的小膠質(zhì)細(xì)胞對(duì)神經(jīng)元細(xì)胞起毒性作用;NBP不能抑制小膠質(zhì)細(xì)胞的激活,但能通過抑制激活的小膠質(zhì)細(xì)胞生成N0,從而減輕激活的小膠質(zhì)細(xì)胞對(duì)神經(jīng)元細(xì)胞的毒性作用,并具有劑量依賴性。
[Abstract]:Objective 1 . To establish and improve the experimental methods and conditions of primary culture of microglial cells in rat cerebral cortex ; 2 . To explore the effect of primary cultured microglial cells on neuronal cells following activation of lipopolysaccharide ( LPS ) in vitro ; 3 . Effects of butylphthalide ( NBP ) on the activation of microglial cells . Methods Primary cultured rat cortical microglial cells and neuronal cells were cultured in primary culture . The cells were stimulated with LPS to stimulate microglial cells . The cell survival rate of neurons was determined by MTT assay . The effects of NBP on the activation of microglial cells were assessed by means of MTT assay . The effects of NBP on the activation of microglial cells were assessed by MTT assay . Results The survival rate of the neurons was decreased after the LPS - activated microglial cells were activated by LPS . The survival rate of the neurons after NBP was decreased , the survival rate of IL - 1尾 , TNF - 偽 and IL - 10 increased , and the survival rate of the neurons after the supernatant was increased compared with the control group , and the above - mentioned effects of butylphthalide showed a dose - dependent manner . Conclusion The activated microglial cells play a toxic role in neuronal cells . NBP does not inhibit the activation of microglial cells , but can inhibit the activation of microglial cells to produce N0 , thus reducing the toxicity of activated microglial cells to neuronal cells and dose - dependent .
