谷跗線(xiàn)螨的形態(tài)學(xué)觀(guān)察、抗原定位及精氨酸激酶基因的克隆與序列分析

【摘要】:由變應(yīng)原引起的變態(tài)反應(yīng)性疾病(如哮喘、變應(yīng)性鼻炎等)是當(dāng)前世界性的重大衛(wèi)生學(xué)問(wèn)題,總發(fā)病率高達(dá)10%~30%,并有逐年增多的趨勢(shì)。谷跗線(xiàn)螨作為一種常見(jiàn)的倉(cāng)儲(chǔ)螨類(lèi),以往人們對(duì)其關(guān)注較少,其形態(tài)特征、組織結(jié)構(gòu)特點(diǎn)和生物學(xué)特性尚不十分明了。 為進(jìn)一步明了谷跗線(xiàn)螨的基本生物學(xué)尤其是其顯微形態(tài)特點(diǎn),探討其與粉塵螨抗原的交叉反應(yīng)性,并嘗試探討其致敏原的相關(guān)分子,本研究進(jìn)行了以下三方面的工作: (1)谷跗線(xiàn)螨的掃描電鏡觀(guān)察及其內(nèi)部主要器官的觀(guān)察:從空調(diào)濾網(wǎng)灰塵中采集谷跗線(xiàn)螨,在體視顯微鏡下清洗處理活螨后,用SEM觀(guān)察其外部形態(tài)特征。經(jīng)形態(tài)鑒定確認(rèn)為該螨種后,以Bouin液固定,制作谷跗線(xiàn)螨石蠟切片,HE染色后光鏡觀(guān)察其內(nèi)部形態(tài)結(jié)構(gòu)。結(jié)果清晰地觀(guān)察到谷跗線(xiàn)螨的體表形態(tài)以及主要的內(nèi)部器官的構(gòu)造,明確了谷跗線(xiàn)螨顎體、背板、基節(jié)板、表皮內(nèi)突、背足體、剛毛等外部形態(tài)以及其消化道等的內(nèi)部結(jié)構(gòu)特點(diǎn)。 (2)谷跗線(xiàn)螨與粉塵螨交叉反應(yīng)性變應(yīng)原的定位研究:制作谷跗線(xiàn)螨石蠟切片,先用對(duì)粉塵螨過(guò)敏患者的陽(yáng)性血清作為一抗孵育,再用生物素標(biāo)記的羊抗人IgE抗體作為二抗,免疫組織化學(xué)染色后在光學(xué)顯微鏡下分析其交叉反應(yīng)性抗原存在位置。研究發(fā)現(xiàn)谷跗線(xiàn)螨口咽部、中腸、腸內(nèi)容物、腸壁及生殖腺等均有陽(yáng)性反應(yīng)。研究表明谷跗線(xiàn)螨體內(nèi)存在可誘發(fā)人體IgE抗體的變應(yīng)原,主要分布于螨消化道、腺體及體壁等部位。 (3)谷跗線(xiàn)螨精氨酸激酶(arginine kinase,AK)基因的克隆及序列分析:從空調(diào)濾網(wǎng)灰塵中采集谷跗線(xiàn)螨(約3000只)提取總RNA,,經(jīng)逆轉(zhuǎn)錄RT-PCR擴(kuò)增谷跗線(xiàn)螨AK基因后測(cè)序?寺~@得的谷跗線(xiàn)螨AK基因由724個(gè)堿基組成,其編碼的蛋白相對(duì)分子質(zhì)量約為26019,等電點(diǎn)為10.10。成功克隆了谷跗線(xiàn)螨AK基因,為進(jìn)行谷跗線(xiàn)螨AK蛋白的重組表達(dá)以及過(guò)敏原性分析奠定基礎(chǔ)。 上述研究積累了有關(guān)谷跗線(xiàn)螨顯微形態(tài)、與已知過(guò)敏原的交叉反應(yīng)性和過(guò)敏原相關(guān)分子研究的新資料,為今后谷跗線(xiàn)螨過(guò)敏性疾病的特異性診斷、治療和預(yù)防奠定工作基礎(chǔ)。
[Abstract]:Allergic diseases ( such as asthma , allergic rhinitis , etc . ) caused by allergens are the major health problems in the world . The total incidence is as high as 10 % -30 % , and there is a tendency to increase year by year . In order to further clarify the basic biology , especially its micro - morphological characteristics , the cross - reactivity with dust mite antigen was discussed , and its related molecules were tried to explore its allergen . The study carried out the following three aspects : ( 1 ) Scanning electron microscope ( SEM ) and observation of main organs were observed . After the mites were collected from the dust of the air - conditioning filter screen , the morphological characteristics of the mites were observed by SEM . ( 2 ) The localization of cross - reactive allergens of tarsomite and dust mites was studied in this paper . The results showed that there were positive responses to the cross - reactive antigens in the mite mouth pharynx , midgut , intestinal contents , intestinal wall and reproductive gland . ( 3 ) Cloning and sequence analysis of the arginine kinase ( AK ) gene of tarsomite : A total RNA was extracted from the dust of the air - conditioning filter screen . The total RNA was amplified by reverse transcription RT - PCR . The AK gene was amplified by RT - PCR . The relative molecular weight of the protein was about 26019 and the isoelectric point was 10.10 . The AK gene was successfully cloned , which laid the foundation for the recombinant expression of AK protein and the analysis of allergen . The above studies have accumulated new information on the micromorphology of tarsonemus , cross - reactivity with known allergens and molecular studies of allergen - related molecules , laying the foundation for the specific diagnosis , treatment and prevention of allergic diseases in the future .
