機(jī)械和感染雙重法建立中華田園犬宮腔粘連模型

【摘要】:目的:通過機(jī)械和感染雙重法構(gòu)建一個(gè)犬的宮腔粘連動(dòng)物模型,并檢測血管內(nèi)皮因子(vascular endothelial growth factor,VEGF)、轉(zhuǎn)化生長因子(transforming growth factor-β1,TGF-β1)的表達(dá),評估其與宮腔粘連形成的關(guān)系,為下一步研究提供實(shí)驗(yàn)平臺。方法:選取6只性發(fā)育成熟的健康雌性中華田園犬,所有建模前的犬的一小段右側(cè)宮角組織作為對照組,建模后該部位子宮角組織作為處理組。利用機(jī)械搔刮和感染雙重法損傷右側(cè)宮角內(nèi)膜,于建模前后各收集一小段宮角標(biāo)本,行HE、Masson染色及SP法免疫組化,整理并統(tǒng)計(jì)分析。結(jié)果:處理組的內(nèi)膜厚度[(2.325±0.399)cm]薄于對照組[(3.025±0.466)cm],差異具有統(tǒng)計(jì)學(xué)意義(P=0.050);處理組的內(nèi)膜腺體數(shù)目[(4.388±1.774)個(gè)]明顯比對照組[(16.518±4.421)個(gè)]減少,差異具有統(tǒng)計(jì)學(xué)意義(P=0.001);處理組的子宮內(nèi)膜纖維化面積比例(0.775±0.040)明顯高于對照組(0.329±0.062),差異有統(tǒng)計(jì)學(xué)意義(P=0.000);處理組的子宮內(nèi)膜中VEGF(5.967±0.390)、TGF-β1(6.200±0.306)的表達(dá)量高于對照組(1.700±0.823)、(1.100±0.361),差異均具有統(tǒng)計(jì)學(xué)意義(P=0.000,P=0.001)。結(jié)論:采用機(jī)械和感染雙重法可以成功構(gòu)建中華田園犬宮腔粘連的動(dòng)物模型,且VEGF、TGF-β1的表達(dá)量升高可能與宮腔粘連的發(fā)生有關(guān)。
[Abstract]:Objective: intrauterine adhesions animal model by mechanical method to build a double infection and canine, and detection of vascular endothelial growth factor (vascular endothelial growth factor VEGF (transforming), transforming growth factor beta 1 beta growth factor-, TGF- 1) expression, and evaluate the relationship between the formation of intrauterine adhesions, provides an experimental platform for the next step of the research. Methods: a total of 6 sexually mature healthy female Chinese pastoral dogs were selected. A small segment of the right cornual tissue of all the dogs before modeling was taken as the control group. After modeling, the uterine horn tissue in this part was taken as the treatment group. The right cornu's intima was damaged by mechanical scraping and infection. A small section of Cornu's specimen was collected before and after modeling. HE, Masson staining and SP immunohistochemical staining were used for statistical analysis. Results: the treatment group endometrial thickness [(2.325 + 0.399) cm] thinner than that of the control group [(3.025 + 0.466) cm], the difference was statistically significant (P=0.050); the treatment group the number of endometrial glands (4.388 + 1.774) [a] was significantly better than the control group [(16.518 + 4.421)] a decreased statistically significant differences (P=0.001); treatment of endometrial fibrosis area ratio group (0.775 + 0.040) was significantly higher than the control group (0.329 + 0.062), the difference was statistically significant (P=0.000); VEGF group of endometrium (5.967 + 0.390), TGF- beta 1 (6.200 + 0.306) expression was higher than that of the control group (1.700 + 0.823), (1.100 + 0.361), the differences were statistically significant (P=0.000, P=0.001). Conclusion: the animal models of intrauterine adhesions in Chinese pastoral dogs can be successfully constructed by dual mechanical and infection methods, and the increased expression of VEGF and TGF- beta 1 may be related to the occurrence of intrauterine adhesions.
