亞麻耐鹽突變體的篩選及種質(zhì)耐鹽性的評(píng)價(jià)

【摘要】:本研究以甲基磺酸乙酯(EMS)處理亞麻萌動(dòng)的種子。根據(jù)M1代的成株率,在纖用亞麻‘范尼’中篩選出最佳的EMS誘變體系,并篩選出具有耐鹽性的突變體以及莖部(株高、莖粗)突變體。并且以來(lái)自國(guó)內(nèi)外的97份亞麻種質(zhì)為研究對(duì)象,運(yùn)用150mmol·L-1NaCl溶液培養(yǎng),以萌發(fā)期種子的發(fā)芽率、發(fā)芽勢(shì)、根長(zhǎng)、苗長(zhǎng)4種生理指標(biāo)變化為依據(jù),利用隸屬函數(shù)法評(píng)價(jià)亞麻萌發(fā)期的耐鹽性,同時(shí)根據(jù)不同種質(zhì)千粒重的變化,分析種子大小與耐鹽性的相關(guān)性。研究結(jié)果如下:1、亞麻突變體庫(kù)的創(chuàng)建及耐鹽突變體的篩選結(jié)果表明:(1)0.6%EMS處理萌動(dòng)亞麻種子,處理時(shí)間為4h;在M1代獲得葉型、葉色、莖型、莖色和株高變化等26種明顯可見(jiàn)性狀突變體,其中M2代能穩(wěn)定遺傳的有17種。在該M2群體中,對(duì)與纖維產(chǎn)量相關(guān)的莖部性狀突變體進(jìn)行了M3代驗(yàn)證,獲得M3代莖部突變體種子。(2)以150mmol·L-1 NaCl溶液篩選M2代,獲得一株耐鹽突變體,其M3代經(jīng)耐鹽性驗(yàn)證獲得M4代。2、97份種質(zhì)耐鹽性鑒定結(jié)果表明:(1)亞麻耐鹽性的評(píng)價(jià)中,各生理指標(biāo)權(quán)重如下:苗長(zhǎng)發(fā)芽勢(shì)根長(zhǎng)發(fā)芽率;(2)相同歐式距離情況下,分配權(quán)重法把97份亞麻種質(zhì)分為5級(jí),算數(shù)平均數(shù)法分為4級(jí);在評(píng)價(jià)作物耐鹽性強(qiáng)弱時(shí),加權(quán)隸屬函數(shù)法能更準(zhǔn)確的衡量品種的耐鹽性;(3)千粒重與亞麻耐鹽性(D值)呈正相關(guān),但相關(guān)系數(shù)較較小;(4)加權(quán)隸屬函數(shù)法把亞麻材料耐鹽性分為Ⅰ—Ⅴ級(jí):Ⅰ級(jí)-2份材料;Ⅱ級(jí)-33份材料;Ⅲ級(jí)-16份材料;Ⅳ級(jí)-28份材料;Ⅴ級(jí)-18份材料。研究補(bǔ)充了前人對(duì)國(guó)內(nèi)外亞麻種質(zhì)耐鹽性評(píng)價(jià)數(shù)據(jù)庫(kù),為今后培育耐鹽性較強(qiáng),農(nóng)藝性狀優(yōu)異亞麻育種工作提供依據(jù)。
[Abstract]:In this study, the germinating seeds of flax were treated with ethyl methanesulfonate (EMS). According to the adult plant rate of M1 generation, the best EMS mutagenesis system was screened from flax 'Fanny', and the salt tolerant mutants and stem (plant height, stem diameter) mutants were screened. 97 flax germplasms from home and abroad were studied and cultured with 150mmol L-1NaCl solution. The changes of germination rate, germination potential, root length and seedling length were taken as the basis. The method of membership function was used to evaluate the salt tolerance of flax during germination, and the correlation between seed size and salt tolerance was analyzed according to the variation of 1000-grain weight of different germplasm. The results were as follows: 1. The establishment of flax mutant bank and the screening of salt tolerant mutants showed that: (1) 0.6%EMS treatment of germinating flax seeds for 4 h; In M1 generation, 26 obvious mutants were obtained, such as leaf type, leaf color, stem type, stem color and plant height change, among which 17 species could be inherited stably in M2 generation. In this M2 population, the mutant of stem traits related to fiber yield was verified by M3 generation, and the seeds of stem mutants of M 3 generation were obtained. (2) A salt tolerant mutant was obtained by screening M2 generation with 150mmol L-1 NaCl solution. The results showed that: (1) in the evaluation of salt tolerance of flax, the weight of each physiological index was as follows: seedling germination rate; (2) under the condition of the same European distance, 97 flax germplasm were divided into five grades by the weight method, the arithmetic average method was divided into 4 grades, and the weighted membership function method could be used to evaluate the salt tolerance of the varieties more accurately when evaluating the salt tolerance of crops.
