Haβ-integrin介導(dǎo)HaCTL3促進(jìn)棉鈴蟲血細(xì)胞包囊反應(yīng)分子機(jī)理研究
[Abstract]:The innate immune system of insects includes humoral and cellular immunity. The immune response mediated by blood cells mainly includes phagocytosis, nodule and cysts, and plays an important role in resisting pathogen infection. The role of cysts is to eliminate intruders whose size is too large to be swallowed by a single blood cell. In this process, the blood cells of insects must change from non-adhesiveness to adhesiveness, and then extend and envelop alien invaders. Our previous study found that a pattern recognition receptor C type lectin 3 (C-type lectin 3H CTL 3) of Helicoverpa armigera can recruit and promote the cysts response of blood cells to gel beads, but the receptor of HaCTL3 on blood cells is not clear. Some studies have shown that 尾 -integrin plays an important role in the process of hemocyte expansion and cysts in insects, but the molecular mechanism is not completely clear. In order to elucidate the function of Ha 尾 -integrin in Helicoverpa armigera and verify whether it plays an important role in the process of promoting hemocyte cysts reaction by HaCTL3, we obtained the gene sequence of Hq 尾 -integrin from the transcription group of Helicoverpa armigera. Ha 尾 -integrin recombinant protein was purified by prokaryotic expression. The expression pattern of Ha 尾 -integrin was analyzed, and the functional experiments related to Ha 尾 -integrin were carried out. The results showed that the expression of 尾 -integrin was highly expressed in the blood cells, and it was mainly located on the membrane of the blood cells. 2Ha 尾 -integrin was involved in the binding of HaCTL3 to the hemocyte membrane, but HaCTL3 could not bind to the surface of the blood cells after the expression of Ha 尾 -integrin. It was suggested that Ha 尾 -integrin might be the plasma membrane receptor of HaCTL3, and that Ha 尾 -integrin and HaCTL3 could interact with each other by co-immunoprecipitation. Knockdown of Ha 尾 -integrin could inhibit the hematopoietic cysts of HaCTL3. In conclusion, we confirm that Ha 尾 -integrin may be the blood membrane receptor of HaCTL3, and play an important role in the process of promoting the cysts of blood cells mediated by HaCTL3. This study will deepen our understanding of the molecular mechanism of hemocyst reaction in insects and provide new clues for biological control of insect pests.
