外源激素對草莓果實(shí)早期發(fā)育及FvCO4基因表達(dá)的影響

【摘要】:果實(shí)的形成和發(fā)育是包括草莓在內(nèi)的瓜果類園藝作物生產(chǎn)的基礎(chǔ)。其中植物激素對果實(shí)發(fā)育和成熟的調(diào)控是果樹學(xué)科研究的重要問題,了解激素與果實(shí)發(fā)育的關(guān)系可為果實(shí)品質(zhì)改良、優(yōu)良品種培養(yǎng)等提供理論參考,也可生為產(chǎn)上應(yīng)用植物激素來改善果實(shí)的品質(zhì)和增加果實(shí)的產(chǎn)量提供依據(jù)。本研究以未授粉材料森林草莓YW5AF7為試材,噴施不同激素于發(fā)育早期的幼果,調(diào)查其對草莓果實(shí)早期發(fā)育的影響,同時(shí)檢測疑似相關(guān)基因FvCO4在草莓果實(shí)髓部和皮層中的表達(dá)變化,為深入了解和闡述外源激素對草莓果實(shí)早期發(fā)育的影響提供試驗(yàn)依據(jù)。主要的研究結(jié)果如下:1.研究了不同濃度的GA_3對森林草莓YW5AF7幼果早期生長發(fā)育的影響。試驗(yàn)發(fā)現(xiàn)采用的不同濃度(400μmol/L、500μmol/L、600μmol/L)GA_3對草莓幼果早期發(fā)育均有促進(jìn)作用,可促進(jìn)草莓果實(shí)髓部和皮層的生長、膨大發(fā)育。其中濃度為500 μmol/L的GA_3作用效果最為顯著。2.分析對比了不同激素(組合)對森林草莓YW5AF7幼果早期生長發(fā)育的影響。結(jié)果表明,相同濃度(500μmol/L)的不同激素處理(GA_3、NAA、NAA+GA_3)均能促進(jìn)草莓幼果早期發(fā)育,其中以500 μmol/L濃度的NAA+GA_3處理影響最為明顯,該處理下幼果發(fā)育大小明顯比同時(shí)期其他兩種激素處理下的幼果發(fā)育更大。3.通過定量RT-PCR的方法檢測了外源激素對草莓果實(shí)中FvCO4基因表達(dá)變化的影響。檢測發(fā)現(xiàn),與對照相比,三種激素處理(GA_3、NAA、NAA+GA_3)均可明顯上調(diào)該基因的表達(dá),其中在幼果形成前期FvCO4的表達(dá)上調(diào)最為明顯。
[Abstract]:Fruit formation and development are the basis for the production of melon and fruit horticultural crops, including strawberries. The regulation of plant hormones on fruit development and maturation is an important issue in fruit tree research. Understanding the relationship between hormone and fruit development can provide theoretical reference for fruit quality improvement and cultivation of fine varieties. It can also provide the basis for the application of plant hormones to improve the quality of fruit and increase the yield of fruit. In this study, unpollinated forest strawberry YW5AF7 was sprayed with different hormones to investigate the effect of YW5AF7 on the early development of strawberry fruit, and the expression of FvCO4 gene was detected in the pulp and cortex of strawberry fruit. To provide experimental basis for further understanding and expounding the effect of exogenous hormones on the early development of strawberry fruit. The main findings are as follows: 1: 1. The effects of different concentrations of GA3 on the early growth and development of young fruits of forest strawberry YW5AF7 were studied. It was found that different concentrations of GA3 (400 渭 mol / L, 500 渭 mol / L, 600 渭 mol / L) could promote the early development of strawberry young fruit, promote the growth of pulp and cortex, and expand development of strawberry fruit. The effect of GA3 at 500 渭 mol / L was the most significant. The effects of different hormones (combinations) on the early growth and development of forest strawberry YW5AF7 young fruit were analyzed and compared. The results showed that different hormone treatments with the same concentration (500 渭 mol / L) could promote the early development of strawberry young fruit, especially 500 渭 mol / L NAA / L NAAGA3 treatment. The developmental size of young fruit under this treatment was significantly larger than that of the other two hormone treatments in the same period. The effects of exogenous hormones on the expression of FvCO4 gene in strawberry fruit were detected by quantitative RT-PCR. The results showed that compared with the control, the expression of FvCO4 was significantly up-regulated by the three hormone treatments (GA3s / NAANAA / GA3), especially in the early stage of young fruit formation.
