高加索三葉草與白三葉雜交后代對(duì)氮和鉀的反應(yīng)及固氮性研究

【摘要】:內(nèi)蒙古大興安嶺林區(qū)分布有逸生型白三葉種群。這個(gè)白三葉種群具有優(yōu)良的環(huán)境適應(yīng)性和固氮能力,已經(jīng)作為父本材料與高加索三葉草雜交,形成大量雜交后代株系。為了提純白三葉根瘤菌用于對(duì)研究材料進(jìn)行接種,觀察雜種F1代接種根瘤菌后的固氮性,同時(shí)試驗(yàn)觀察氮鉀元素對(duì)F1代株系生長的影響,本試驗(yàn)首先對(duì)白三葉根瘤菌培養(yǎng)條件(培養(yǎng)基初始pH值、培養(yǎng)溫度和搖床轉(zhuǎn)速)進(jìn)行了篩選,提純了根瘤菌。根瘤菌培養(yǎng)條件采用單因素和雙因素設(shè)計(jì),3次重復(fù);氮鉀元素處理采用雙因素設(shè)計(jì),3次重復(fù),分別對(duì)F1代株系及親本進(jìn)行不同施氮和施鉀水平處理,觀察不同氮鉀水平處理下對(duì)植株的生理指標(biāo)與根系的影響;對(duì)F1代株系及2種親本進(jìn)行根瘤菌接種試驗(yàn),將發(fā)芽后的種子移栽至滅菌后的蛭石中,分別進(jìn)行不接種根瘤菌、接種根瘤菌激活和接種根瘤菌不激活3種處理,3次重復(fù),觀察3種處理對(duì)植株根系及干重的影響,并用元素分析儀測(cè)定植株的含氮量,測(cè)定是否具有固氮能力。結(jié)果表明:1.在單因素處理?xiàng)l件下,白三葉根瘤菌的理想培養(yǎng)條件是培養(yǎng)基初始pH值為7.0,培養(yǎng)溫度為25℃,搖床轉(zhuǎn)速為180r/min。在雙因素處理?xiàng)l件下,當(dāng)溫度和初始pH值為自變量時(shí),溫度為25℃、pH值為7.0時(shí),適宜根瘤菌生長;在轉(zhuǎn)速與初始pH值為自變量時(shí),轉(zhuǎn)速為160r/min、pH值為7.0時(shí),適宜根瘤菌生長;當(dāng)轉(zhuǎn)速與溫度為自變量時(shí),轉(zhuǎn)速和溫度分別為160r/min和25℃時(shí),適合根瘤菌生長。2.在不同氮鉀元素處理下,植株的生理指標(biāo)和根系指標(biāo)會(huì)隨著施氮量與施鉀量的增加先增加后降低,在氮鉀元素處理下,植株表現(xiàn)為氮為主要影響因素,親本對(duì)氮鉀元素的反應(yīng)更為強(qiáng)烈。3.隨著接種根瘤菌,白三葉根系及干重指標(biāo)明顯升高;在接種根瘤菌處理下,F1代株系及高加索三葉草仍表現(xiàn)為固氮性缺失,白三葉在接種根瘤菌后固氮性增強(qiáng)。
[Abstract]:In Daxingan Mountains, Inner Mongolia, there is an ectogenetic white clover population. This white clover population has excellent environmental adaptability and nitrogen fixation ability and has been used as a male parent to cross with Caucasian clover to form a large number of hybrid progeny lines. In order to purify white clover rhizobia for inoculation, the nitrogen-fixation of F1 hybrids was observed, and the effects of nitrogen and potassium on the growth of F1 strains were also observed. In this experiment, the culture conditions (initial pH value of culture medium, culture temperature and shaking table speed) were screened and the rhizobia were purified. The culture conditions of rhizobia were designed three times by single factor and two factors, and the treatments of nitrogen and potassium were used to treat F1 strains and their parents with different levels of nitrogen and potassium respectively. The effects of different nitrogen and potassium levels on plant physiological index and root system were observed, and rhizobia inoculation test was carried out on F1 strain and two parents. The germinated seeds were transplanted into sterilized vermiculite and inoculated with rhizobia respectively. The effects of three treatments on the root system and dry weight of the plants were observed. The nitrogen content of the plants was determined by elemental analyzer and the ability of nitrogen fixation was determined. The result shows that 1: 1.
