空心蓮子草對蓮草直胸跳甲取食脅迫的生理生化響應(yīng)

【摘要】:植物在與植食性昆蟲的協(xié)同進(jìn)化過程中,逐漸形成了完善的防御措施來應(yīng)對昆蟲的取食?招纳徸硬葑鳛橐环N外來入侵雜草,分布廣泛,危害嚴(yán)重,其專食性天敵蓮草直胸跳甲則被廣泛用于空心蓮子草的防治當(dāng)中。本研究主要以空心蓮子草應(yīng)對蓮草直胸跳甲的取食脅迫后空心蓮子草葉片內(nèi)防御酶、脫落酸以及空心蓮子草揮發(fā)物的變化來探究空心蓮子草的防御機制。通過對空心蓮子草進(jìn)行剪葉及蓮草直胸跳甲取食處理24 h后,測定葉片內(nèi)防御酶活性(PPO、POD、LOX和PAL),發(fā)現(xiàn)各防御酶活性變化并不完全相同,且各防御酶活性與處理時間及處理方式有著密切的關(guān)系。采用動態(tài)頂空收集法,對經(jīng)不同蟲態(tài)蓮草直胸跳甲取食后空心蓮子草的揮發(fā)物進(jìn)行采集分析。結(jié)果表明,空心蓮子草經(jīng)不同蟲態(tài)蓮草直胸跳甲取食后收集到的揮發(fā)物數(shù)量與含量成正相關(guān)的關(guān)系?招纳徸硬萁(jīng)1齡幼蟲、3齡幼蟲及成蟲取食后揮發(fā)性物質(zhì)種類數(shù)量分別為45種、16種和6種,而對照組空心蓮子草揮發(fā)物共捕獲8種。對經(jīng)蓮草直胸跳甲1齡幼蟲、3齡幼蟲及成蟲持續(xù)取食24 h的空心蓮子草葉片ABA含量進(jìn)行測定發(fā)現(xiàn),各處理后空心蓮子草葉片內(nèi)脫落酸ABA含量均高于對照組,其中1齡幼蟲和3齡幼蟲顯著高于對照組。綜上所述,機械損傷及昆蟲取食均能誘導(dǎo)空心蓮子草各防御酶活性的升高;被蓮草直胸跳甲取食后的空心蓮子草揮發(fā)物發(fā)生改變,并且葉片內(nèi)ABA含量升高,說明空心蓮子草經(jīng)機械損傷和蓮草直胸跳甲取食脅迫后可產(chǎn)生一定的防御反應(yīng),但是其防御機制(如葉片內(nèi)營養(yǎng)物質(zhì)含量、次生代謝產(chǎn)物的變化等)及各防御功能之間的關(guān)系還有待進(jìn)一步研究。
[Abstract]:In the process of co-evolution between plants and phytophagous insects, perfect defense measures were gradually formed to deal with insect feeding. As an alien invasive weed, Lotus seed is widely distributed and seriously damaged, and its special natural enemy, Lilium chinensis, is widely used in the prevention and control of lotus seed. In this study, the defense mechanism of Hollow Lotus seed was studied by the changes of defense enzymes, abscisic acid and volatile matter in leaves after feeding stress. The defense enzyme activity of PPOP POD LOX and PALLX in leaves were determined after 24 hours of leaf clipping and straight breast armour feeding on lotus seed. The results showed that the changes of defense enzyme activities were not exactly the same in leaves, and the results showed that the changes of defense enzyme activity were not the same as those of PPOP PODX and PALX. The activity of each defense enzyme was closely related to the treatment time and treatment mode. Dynamic headspace collection method was used to collect and analyze the volatiles of lotus seed after feeding with different insect species. The results showed that there was a positive correlation between the amount of volatiles collected by different insect species and the content of the volatile compounds. The number of volatile substances in the third instar larvae and adults were 45 species, 16 species and 6 species, respectively, while in the control group, 8 species were captured. The content of ABA in the leaves of the third instar larvae and adults fed on the first instar larvae of Lilifera chinensis for 24 h was determined. The results showed that the content of abscisic acid ABA in the leaves of each treatment was higher than that in the control group. The 1st instar larva and 3rd instar larva were significantly higher than the control group. In conclusion, mechanical injury and insect feeding could induce the increase of the activities of defense enzymes, and the changes of the volatiles and the increase of ABA content in leaves. The results showed that lotus seed could produce a certain defense response after mechanical injury and feeding stress, but its defense mechanism (such as the content of nutrients in leaves), The changes of secondary metabolites, etc.) and the relationship between each defense function need to be further studied.
