蒙古口蘑菌絲體生長(zhǎng)發(fā)育及其漆酶表達(dá)特性研究

【摘要】:蒙古口蘑是我國(guó)北方草原珍貴的食藥用菌,同時(shí)作為草原生物鏈的重要環(huán)節(jié),其豐富均衡的營(yíng)養(yǎng)與多樣的藥用成分,使其具有極高的利用價(jià)值和開發(fā)前景。目前,蒙古口蘑已被列入《國(guó)家重點(diǎn)保護(hù)野生植物名錄(第二批)》,為使這一珍貴的種質(zhì)資源得到有效保護(hù),加強(qiáng)研究其生理學(xué)及馴化栽培技術(shù)十分必要。本試驗(yàn)研究了蒙古口蘑擔(dān)孢子萌發(fā)及其初生菌絲生物學(xué)特性,鑒定了蒙古口蘑擔(dān)孢子交配類型,測(cè)定了蒙古口蘑菌絲體漆酶活性,研究了低溫刺激對(duì)蒙古口蘑二次菌絲漆酶基因表達(dá)的影響。以期通過本試驗(yàn),積累蒙古口蘑人工栽培等基礎(chǔ)性研究的資料。本論文主要結(jié)論如下:1、蒙古口蘑擔(dān)孢子無色,腎形或橢圓形,表面光滑,擔(dān)孢子大小約為(4~6)μm×(6~9)μm,可被齊氏(Ziehl)石炭酸復(fù)紅染液染色;擔(dān)孢子在PDA及MS固體培養(yǎng)基上,25℃黑暗條件下至少一周的時(shí)間才能萌發(fā)出肉眼可見的微小菌落;MS培養(yǎng)基中添加1.5g/L酵母粉和1.5g/L水解酪蛋白,對(duì)蒙古口蘑擔(dān)孢子萌發(fā)和初生菌絲生長(zhǎng)有明顯的促進(jìn)作用。2、根據(jù)菌落生長(zhǎng)速度和菌落形態(tài)等有關(guān)特征,可把初生菌絲分為五種類型,分別為菌絲絨毛型、菌絲貼生型、菌絲稀薄型、成簇生長(zhǎng)型、緩慢生長(zhǎng)型。3、蒙古口蘑交配類型屬于四極性交配系統(tǒng),為A、B因子互不連鎖的雙因子控制系統(tǒng)。可觀察到典型的四種交配反應(yīng)菌落形態(tài):親和反應(yīng)、柵欄反應(yīng)、平貼反應(yīng)、無交界不親和反應(yīng)。4、以蒙古口蘑的次生菌絲體為材料,研究低溫刺激對(duì)蒙古口蘑漆酶活力及漆酶基因家族轉(zhuǎn)錄的影響:1)蒙古口蘑不同菌株,漆酶活力不同,且隨菌絲體的不斷生長(zhǎng),漆酶活性變化較大,總體呈下降趨勢(shì)。2)低溫刺激可不同程度地提高或降低蒙古口蘑漆酶基因家族的轉(zhuǎn)錄水平,5℃刺激可抑制漆酶基因家族轉(zhuǎn)錄水平,10℃刺激可提高漆酶基因家族轉(zhuǎn)錄水平,15℃刺激也可使漆酶基因家族轉(zhuǎn)錄水平明顯變化,整體上隨著處理溫度的提高,呈先升高再降低的趨勢(shì)。所有處理?xiàng)l件中,以10℃處理10天蒙古口蘑漆酶活力提高最顯著。
[Abstract]:Tricholoma mongolicus is a precious edible and medicinal fungus in northern grassland of China. As an important link of grassland biological chain, it is rich in balanced nutrition and various medicinal components, which makes it have high utilization value and development prospect. At present, Tricholoma mongolica has been listed in the list of National key protected Wild plants (the second batch). In order to protect this precious germplasm resource effectively, it is necessary to study its physiology and domestication techniques. In this paper, the germination and the biological characteristics of the primary hyphae of Tricholoma mongolica were studied, and the mating type and laccase activity of the mycelium of Tricholoma mongolica were identified. The effects of low temperature stimulation on the expression of laccase gene in secondary mycelia of Tricholoma mongolica were studied. In order to accumulate the data of the basic research such as artificial cultivation of Tricholoma mongolica through this experiment.
