齒瓣石斛多糖提取工藝及抗氧化活性研究

【摘要】:如今,現(xiàn)代園林在觀賞游憩的基礎(chǔ)上,更加注重其生態(tài)功能,為周圍小環(huán)境的改善發(fā)揮了重要作用,同時,維護(hù)公共健康也成為園林所擔(dān)負(fù)的重要責(zé)任。作為構(gòu)建養(yǎng)生保健型園林的基礎(chǔ),將具有保健作用的藥用園林植物引入現(xiàn)代園林成為了時尚。本研究就是基于此,對藥用園林植物中眾多種類中的一種——齒瓣石斛(Dendrobium devonianum Paxt.)進(jìn)行深入探究,目的是使其在充分保證園林生態(tài)功能和美化功能的前提下,為進(jìn)一步開發(fā)其醫(yī)療保健功能提供科學(xué)依據(jù)和理論指導(dǎo)。齒瓣石斛作為石斛屬的一種,對其研究還大多在栽培種植上,對于其自身代謝產(chǎn)物的研究還是空白,因此,對齒瓣石斛多糖的酶提取工藝研究和抗氧化活性研究具有重要的意義。本實驗首先對齒瓣石斛多糖的酶提法進(jìn)行了工藝優(yōu)化,通過單因素實驗確定對其提取率影響最大的三個因素為酶量濃度、酶解溫度和pH。然后通過Box-Behnken響應(yīng)面實驗對其提取工藝進(jìn)行優(yōu)化,優(yōu)化后的最佳提取條件為:酶量濃度0.94%,酶解溫度51.06℃,pH=4.94。在該條件下,預(yù)測的齒瓣石斛多糖提取率為157.93 mg/g。經(jīng)3次驗證實驗,測得其平均提取率為157.58 mg/g,與預(yù)測值基本吻合。然后根據(jù)提取工藝的方法,對齒瓣石斛多糖進(jìn)行提取。將提取液用sevage試劑去蛋白,檢測其在280 nm處的紫外吸收峰判斷溶液中是否還含有蛋白質(zhì)。去蛋白后,將多糖提取液反復(fù)醇沉以去除色素,直到提取液顏色接近無色時,分別在乙醇濃度為40%、60%和80%下醇沉出三種不同分子量的多糖,并將其命名為40%多糖、60%多糖和80%多糖。將三種多糖用丙酮和乙酸乙酯洗滌以去除有機(jī)雜質(zhì)后,再次溶解,置于透析袋中透析以去除小分子雜質(zhì)。最后將透析后的多糖溶液減壓干燥,得到固體多糖。最后將三種多糖進(jìn)行體外抗氧化活性的測定。結(jié)果顯示,通過酶提法提取出來的齒瓣石斛多糖抗氧化活性很低。40%多糖、60%多糖和80%多糖的糖醛酸含量分別僅為3.05%、2.96%和2.98%,硫酸根含量分別僅為0、0.70%和O。判定還原性大小的還原值僅分別為0.051、0.011和0.021,對超氧自由基的清除率分別為5.63%、9.22%、5.63%,對DPPH自由基的清除率分別為0、0和1.43%,對ABTS自由基的清除率分別為0.37%、0.03%和14.86%,對羥自由基的清除率分別為27.54%、0.70%和0。
[Abstract]:Nowadays, modern gardens pay more attention to their ecological function and play an important role in improving the surrounding environment on the basis of viewing and recreation. At the same time, the maintenance of public health has also become an important responsibility of gardens. As the foundation of constructing health care garden, it has become fashionable to introduce medicinal garden plants with health care function into modern garden. Based on this, a new species of medicinal garden plant, Dendrobium devonianum Paxt.was studied. The purpose of this study is to provide scientific basis and theoretical guidance for the further development of its medical and health care function on the premise of fully guaranteeing the ecological function and beautifying function of the garden. As a species of Dendrobium, the research on Dendrobium odontoides is mostly in cultivation and planting, and the research on its own metabolites is still blank. It is of great significance to study the enzymatic extraction and antioxidant activity of Dendrobium candidum polysaccharides. In this experiment, the enzymatic extraction of Dendrobium Dendrobium polysaccharides was optimized. The three factors that had the greatest influence on the extraction rate of Dendrobium Dendrobium were the concentration of enzyme, the temperature of enzymatic hydrolysis and the pH of Dendrobium dendrobium. Then the extraction process was optimized by Box-Behnken response surface experiment.
