鱗頭犬牙南極魚卵殼蛋白的功能初步研究

【摘要】:已有研究表明卵殼蛋白ZPC5(Dissostichus mawsoni zona pellucida c5,Dm ZPC5)在南極魚卵巢中大量表達(dá),但是其具體功能尚不清楚。為了明確ZPC5是否參與魚類抗寒過程,研究在中國倉鼠卵巢細(xì)胞(Chinese hamster ovary,CHO)中過表達(dá)南極魚ZPC5,隨之給予CHO細(xì)胞一定程度的低溫處理,通過統(tǒng)計(jì)CHO細(xì)胞的存活率,來探討zpc5在魚類抗寒過程中的作用。存活率檢測(cè)結(jié)果發(fā)現(xiàn),過表達(dá)ZPC5的CHO細(xì)胞在0℃處理16 h后,其存活率較對(duì)照組顯著上升(P0.001),證明zpc5在低溫下可以促進(jìn)細(xì)胞的存活。Western Blot檢測(cè)結(jié)果顯示,zpc5能在CHO細(xì)胞中高效表達(dá),0℃處理后,CHO細(xì)胞內(nèi)的ZPC5表達(dá)量顯著上調(diào),證明了低溫可以誘導(dǎo)ZPC5的表達(dá)。研究結(jié)果表明,ZPC5有可能參與南極魚魚卵抗寒過程。
