平菇優(yōu)良菌株的選育及評價

【摘要】:本試驗收集了冀中南地區(qū)33個平菇栽培菌株,通過酯酶同工酶、ISSR分子標(biāo)記分析、拮抗試驗以及出菇試驗,對33個平菇栽培菌株進(jìn)行了生理生化和分子標(biāo)記鑒定。在此基礎(chǔ)上,選取了生產(chǎn)性狀優(yōu)劣互補(bǔ)的“99”、“680”和“雙抗”三個菌株作為親本。通過單孢雜交配對和原生質(zhì)體融合,共獲得176個雜交子代菌株,并對其進(jìn)行一些列篩選試驗,旨在獲得平菇優(yōu)良菌株。1.通過拮抗測定、ISSR分子生物標(biāo)記分析和酯酶同工酶酶譜為依據(jù),將供試菌株分為7大類,其中栽培量最大的是以“89”、“99”和“雙抗黑平”為代表的三大類菌株。2.通過單孢分離技術(shù)分離了三個菌株的擔(dān)孢子,得到“99”單核體2個,“680”單核體5個,“雙抗”單核體3個。選取菌絲生長較快的親本“99”和“680”單核菌絲分別與“雙抗”單核菌絲進(jìn)行了兩兩雜交配對,得到21個雜交組合菌株。3.根據(jù)鎖狀聯(lián)合有無進(jìn)一步確定雜交子,篩選出了10個優(yōu)良雜交子,采用拮抗、酯酶同工酶及出菇等方法對融合子進(jìn)行了進(jìn)一步鑒定和評價分析。10個雜交子均與親本有明顯的拮抗線,且具有兩親本菌株的典型酯酶同工酶條帶,確定是雜交菌株,以此為依據(jù)進(jìn)一步篩選出了4個平菇優(yōu)良雜交子,都具備出菇能力,分別編號為“Pz945”,“Pz935”,“Pz675”,“Pz6115”。4.同時制備平菇優(yōu)良品種“99”和“雙抗”原生質(zhì)體,并分離了單核體進(jìn)行融合。得到融合子155個,根據(jù)鎖狀聯(lián)合有無進(jìn)一步確定融合子,通過菌絲長勢篩選出14個長勢較好的融合。
[Abstract]:In this experiment, 33 cultivated strains of Pleurotus ostreatus were collected in the middle and south of Hebei Province. The physiological, biochemical and molecular markers of 33 cultivated strains of Pleurotus ostreatus were identified by ISSR molecular marker analysis, antagonistic test and mushroom production test. On the basis of this, 33 cultivated strains of Pleurotus ostreatus were identified by physiological, biochemical and molecular markers. Three strains of "99", "680" and "double resistance" with complementary production traits were selected as parents. 176 hybrid progenies were obtained by single sporulation pairing and protoplast fusion, and some screening tests were carried out on them. The purpose of this study was to obtain fine strains of Pleurotus ostreatus. Based on the analysis of ISSR molecular biomarkers and esterase isozyme patterns, the tested strains were divided into 7 groups. Among them, the most cultivated strains were "89", "99" and "double resistance to black flat". Two monokaryons of "99", five monosomes of "680" and three monosomes of "double antibody" were obtained. 21 hybrid combination strains. 3. According to the combination of lock-like combination to further determine the hybrids, 10 excellent hybrids were screened, and antagonistic methods were used. The fusion was further identified and evaluated by esterase isozyme and mushroom extraction methods. The 10 hybrids had obvious antagonistic lines with their parents, and the typical esterase isozyme bands of two parent strains were identified as hybrid strains. On this basis, four fine hybrids of Pleurotus ostreatus were further selected, all of which had the ability to produce Pleurotus ostreatus, numbered "Pz945", "Pz935", "Pz675", "Pz6115" .4.At the same time, the protoplasts of "99" and "double resistance" of Pleurotus ostreatus were prepared. According to the combination of lock-like or not, 14 fusants with good growth were screened by hyphal growth.
