磺胺氯吡嗪鈉-二甲氧芐啶可溶性粉在雞體內(nèi)的藥代動(dòng)力學(xué)研究

【摘要】:磺胺氯吡嗪鈉-二甲氧芐啶可溶性粉是一種新型抗球蟲(chóng)的復(fù)方制劑,它可以清除由柔嫩、布氏、毒害等多種艾美耳球蟲(chóng)引起的雞的腸道感染,同時(shí)對(duì)家兔、羔羊的球蟲(chóng)病也具有很好的療效;前仿冗拎衡c主要作用于球蟲(chóng)的活性峰期的第二代裂殖體,而且它對(duì)第一代裂殖體也起到一些作用。它主要用于球蟲(chóng)的爆發(fā)短期使用。它還具有抗菌作用,也可以治療禽霍亂及雞傷寒。二甲氧芐啶作為一種磺胺藥物的增效劑,也具有抗菌作用。目前來(lái)看,它是一種很有效的抗球蟲(chóng)病的復(fù)方制劑,在抗雞球蟲(chóng)病的過(guò)程中起了很重要的作用。在世界范圍內(nèi),它的使用很廣,尤其在美國(guó)、歐洲等地。它便于儲(chǔ)存、運(yùn)輸,但在國(guó)內(nèi)還未上市。本研究建立高效液相的檢測(cè)方法,并完成磺胺氯吡嗪鈉-二甲氧芐啶可溶性粉在雞體內(nèi)的藥代動(dòng)力學(xué)研究。高效液相檢測(cè)的色譜條件:色譜柱為大連伊力特色譜柱(250*4.6mm,5μm),填料為Hypersil ODS2,以0.1%甲酸水-乙腈(V/V=70:30)為流動(dòng)相,流速為1m L/min,柱溫為30℃,進(jìn)樣量為10μL。血漿樣品使用乙腈做前處理后,制備待檢測(cè)樣品;前仿冗拎衡c在0.1μg/m L—40μg/m L線性關(guān)系良好,二甲氧芐啶在0.1μg/m L—4μg/m L線性關(guān)系良好,該方法磺胺氯吡嗪鈉、二甲氧芐啶的檢測(cè)限均為0.05μg/m L,定量限為0.1μg/m L,該方法磺胺氯吡嗪鈉的平均回收率為85.40%-97.13%,二甲氧芐啶的平均回收率為86%-97.36%,日內(nèi)與日間變異系數(shù)均小于15%。且磺胺氯吡嗪鈉、二甲氧芐啶在室溫、三次反復(fù)凍融、-80℃冷凍條件下的穩(wěn)定性均良好,符合生物樣品的分析要求。并且該方法的靈敏度高,操作簡(jiǎn)便,可以用于磺胺氯吡嗪鈉-二甲氧芐啶可溶性粉在雞體內(nèi)的藥代動(dòng)力學(xué)研究。藥代動(dòng)力學(xué)結(jié)果:通過(guò)給白羽肉雞灌服低、中、高三種不同濃度的藥物,使用DAS 3.0軟件非房室模型對(duì)數(shù)據(jù)進(jìn)行計(jì)算。兩種藥物的藥代動(dòng)力學(xué)參數(shù)如下:磺胺氯吡嗪鈉的達(dá)峰時(shí)間T_(max)分別為7.833±1.337、9.333±1.303、9.667±1.155h;峰濃度C_(max)分別為10.16±0.922、17.457±0.663、26.905±2.154μg/m L;半衰期t_(1/2)分別為21.303±9.034、22.027±5.568、15.405±1.953h;藥時(shí)曲線下面積AUC分別為288.534±43.743、568.828±56.072、859.145±90.379μg/m L*h。二甲氧芐啶的達(dá)峰時(shí)間T_(max)分別為4±0.853、4.333±1.435、4.333±1.435h;峰濃度C_(max)分別為0.261±0.045、0.37±0.065、0.66±0.059μg/m L;半衰期t_(1/2)分別為12.836±16.301、19.728±8.689、16.774±3.824h;藥時(shí)曲線下面積AUC分別為2.013±0.867、5.383±1.882、11.212±3.7μg/m L*h。以上數(shù)據(jù)為磺胺氯吡嗪鈉-二甲氧芐啶在臨床的合理用藥提供了理論依據(jù)。
[Abstract]:Sulfamethopyrazine-dimethoprim soluble powder is a new compound preparation against coccidiosis. It can eliminate the intestinal tract infection of chicken caused by Eimeria tenella, Brucella, toxicity and so on, and at the same time, it can treat rabbits. Sodium sulfamethopyrazine mainly acts on the second generation merozoite of coccidiosis in the peak stage of the activity of coccidiosis. And it also has some effect on the first generation of merozoites. It is mainly used for short-term use of coccidiosis. It also has antibacterial effect. Dimethoprim, as a synergist of sulfonamides, also has antimicrobial effect. At present, it is a very effective compound preparation against coccidiosis. It plays an important role in the fight against coccidiosis. It is widely used in the world, especially in the United States, Europe and other places. It is easy to store and transport. However, it has not been listed in China. In this study, a method for the detection of high performance liquid phase (HPLC) was established. The pharmacokinetics of sulfamethopyrazine-dimethoprim soluble powder in chicken was studied. The chromatographic conditions of high performance liquid chromatography (HPLC) were as follows: Dalian Eliot column 2504.6mm.
