高效復(fù)合微生態(tài)制劑的研制及其應(yīng)用

【摘要】:微生態(tài)制劑是一種替代抗生素的新型產(chǎn)品,以其無殘留、無耐藥性、無毒副作用、效果顯著等優(yōu)點(diǎn)廣泛應(yīng)用在水產(chǎn)養(yǎng)殖、動(dòng)物養(yǎng)殖、種植業(yè)以及疾病治療等領(lǐng)域。本研究的內(nèi)容主要在于篩選新的菌株,制備復(fù)合微生態(tài)制劑以及針對(duì)性地應(yīng)用于水產(chǎn)養(yǎng)殖水處理。具體研究?jī)?nèi)容包括:1、從滄州和天津地區(qū)采集并篩選出8個(gè)有效底泥樣品,利用選擇培養(yǎng)基篩選出了11株特性菌株。進(jìn)而對(duì)篩選得到菌株進(jìn)行了初步性能檢測(cè),得到4株處理人工廢水最佳的菌株,對(duì)它們分別編號(hào)為21-B、31-A、33-A、41-A。2、通過分子實(shí)驗(yàn)和生理生化實(shí)驗(yàn)對(duì)篩選的菌株進(jìn)行鑒定,鑒定結(jié)果是21-B菌為阪崎腸桿菌(Cronobacter sakazakii),31-A菌為枯草芽孢桿菌(Bacillus subtilis),33-A菌為球形賴氨酸芽孢桿菌(Lysinibacillus sphaericus),41-A菌為蠟狀芽孢桿菌(Bacillus cereus)。3、單獨(dú)對(duì)阪崎腸桿菌進(jìn)行脫氮性能檢測(cè),該菌株對(duì)亞硝酸鹽的去除率為97.6%,對(duì)氨氮的去除率高達(dá)98.3%。4、將得到的另外3株單菌株進(jìn)行混合培養(yǎng)。優(yōu)化的混合培養(yǎng)基為葡萄糖10 g?L-1、酪蛋白10 g?L-1、磷酸氫二鉀2 g?L-1和牛肝浸粉1 g?L-1;最適培養(yǎng)溫度為37℃,最適生長(zhǎng)pH為8.0,最適的轉(zhuǎn)速為120 r?min-1。最優(yōu)的復(fù)合比是31-A:33-A:41-A為2:1:1,即7號(hào)組合處理廢水的效果最佳。5、將7號(hào)混合菌制劑進(jìn)行應(yīng)用實(shí)驗(yàn)。其中在模擬性能檢測(cè)中,亞硝酸鹽的去除率達(dá)75%,氨氮的去除率在40%。在采集水實(shí)驗(yàn)中驗(yàn)證得出混合菌劑對(duì)水體中COD的去除率在30%左右,并且證明處理的最適條件與菌株最適的生長(zhǎng)條件一致。
[Abstract]:Probiotics is a new product to replace antibiotics, with no residue, no resistance, no toxic and side effects, the advantages of widely used in aquaculture, animal breeding, planting and treatment of diseases and other fields. The contents of the research mainly lies in the screening of new strains of probiotics and preparation for the application of water treatment in aquaculture. The research contents include: 1, collected from Cangzhou and Tianjin area and screened 8 sediment samples, using selective medium were screened from 11 strains of characteristic strain. And made a primary performance test of screened strains, 4 strains of artificial wastewater processing strains best. They were named 21-B, 31-A, 33-A, 41-A.2, through experiment and physiological and biochemical experiments in molecular identification of strains, the identification results are 21-B strains of Enterobacter sakazakii (Cronobacter sakazakii), 31- The strain A was Bacillus subtilis (Bacillus subtilis), 33-A acid bacteria Bacillus Lai (Lysinibacillus sphaericus), spherical 41-A strains of Bacillus cereus (Bacillus cereus).3, of Enterobacter sakazakii in nitrogen removal performance test, the strain on the nitrite removal rate was 97.6%, the removal of ammonia the rate of up to 98.3%.4, the other 3 were single strains were mixed culture. The optimization of mixed medium was glucose 10 g? L-1, G L-1, 10 casein? Two potassium hydrogen phosphate 2 g? L-1 and bovine liver extract powder 1 g? Min-1. optimal composite ratio of 31-A:33-A:41-A to 2:1:1, 7 combined wastewater treatment effect is the best.5, No. 7 mixed bacteria application in simulation experiment. Performance test, the nitrite removal rate was 75%, the removal rate of ammonia nitrogen in 40%. collection The water experiment showed that the removal rate of COD in the water body was about 30%, and the optimum conditions for the treatment were the same as the optimum conditions for the strain.
