NLRX1和Sam68調(diào)控丙型肝炎病毒復(fù)制機制研究

丙型肝炎病毒(HCV)是一種重要的人類病原體,全世界有超過1.7億人被感染,它可導(dǎo)致慢性肝炎、肝硬化、甚至肝癌。目前還沒有有效針對丙型肝炎病毒的疫苗。HCV是一種單股正鏈RNA病毒,編碼產(chǎn)生3種結(jié)構(gòu)蛋白和7種非結(jié)構(gòu)蛋白。HCV感染人類細(xì)胞后,受到宿主先天免疫應(yīng)答的調(diào)控。病毒入侵宿主細(xì)胞后,模式識別受體RIG-I樣受體識別病毒的病原相關(guān)分子模式(PAMPs),RIG-I和MDA5被激活后發(fā)生構(gòu)象變化,激活定位在線粒體外膜上的接頭蛋白MAVS,MAVS招募并激活I(lǐng)RF3和NF-kB,最終誘導(dǎo)I型、III型干擾素以及其他抗病毒細(xì)胞因子的產(chǎn)生。我們的研究發(fā)現(xiàn),HCV上調(diào)肝細(xì)胞內(nèi)的NLRX1蛋白水平。沉默NLRX1可抑制HCV復(fù)制,而過表達NLRX1則促進HCV復(fù)制和病毒蛋白的積累。在穩(wěn)定沉默NLRX1的細(xì)胞系中感染HCV,I型干擾素、III型干擾素和干擾素誘導(dǎo)基因(ISGs)的mRNA水平都明顯上升。進一步研究發(fā)現(xiàn),NLRX1與接頭蛋白MAVS結(jié)合,誘導(dǎo)MAVS泛素化降解。深入研究發(fā)現(xiàn),NLRX1與PCBP2相互作用,并促進MAVS的K48位泛素化。除了HCV之外,NLRX1對其它RNA病毒誘...
