人參皂苷Rg3抑制人骨肉瘤細胞增殖及其分子機制研究

研究背景:骨肉瘤是發(fā)源于間充質干細胞的惡性腫瘤。骨肉瘤的發(fā)病年齡呈現(xiàn)雙峰分布,常發(fā)病于青少年或老年人。由于大多數(shù)的骨肉瘤患者在診斷時時就有轉移或微轉移性,故幾乎所有患者在手術切除的之前或之后都會接受多種藥物聯(lián)合化療。患者往往耐受度較差,且復發(fā)幾率較高。人參是一種原產于東北亞的草本植物,在東亞地區(qū)的傳統(tǒng)醫(yī)學中有著廣泛的應用。作為人參中主要的活性物質之一,人參皂苷Rg3的抗腫瘤作用在近年來越來越引起人們的重視。人參皂苷Rg3的甾體結構使得它可以與細胞膜、膜結合離子通道、以及胞內和胞外的受體相互作用來改變細胞的轉錄水平以來發(fā)揮抗腫瘤作用。研究方法:本實驗應用MTT實驗、流式細胞術、細胞劃痕實驗、Transwell細胞侵襲實驗檢測了人參皂苷Rg3對骨肉瘤細胞系MG-63、HOS和U2OS增殖、凋亡、遷移、侵襲的影響和對細胞周期作用。我們又進一步選擇MG-63細胞作為研究對象,應用western blot、定量PCR、流式細胞術、小干擾RNA等實驗手段進一步探究人參皂苷Rg3誘導細胞自噬、凋亡以及細胞周期阻滯的具體作用機制。實驗結果:MTT實驗發(fā)現(xiàn)人參皂苷Rg3以劑量依賴的方式抑制三種骨肉瘤細胞...
