人參皂苷Rg3抑制人骨肉瘤細(xì)胞增殖及其分子機(jī)制研究

研究背景:骨肉瘤是發(fā)源于間充質(zhì)干細(xì)胞的惡性腫瘤。骨肉瘤的發(fā)病年齡呈現(xiàn)雙峰分布,常發(fā)病于青少年或老年人。由于大多數(shù)的骨肉瘤患者在診斷時(shí)時(shí)就有轉(zhuǎn)移或微轉(zhuǎn)移性,故幾乎所有患者在手術(shù)切除的之前或之后都會(huì)接受多種藥物聯(lián)合化療;颊咄褪芏容^差,且復(fù)發(fā)幾率較高。人參是一種原產(chǎn)于東北亞的草本植物,在東亞地區(qū)的傳統(tǒng)醫(yī)學(xué)中有著廣泛的應(yīng)用。作為人參中主要的活性物質(zhì)之一,人參皂苷Rg3的抗腫瘤作用在近年來越來越引起人們的重視。人參皂苷Rg3的甾體結(jié)構(gòu)使得它可以與細(xì)胞膜、膜結(jié)合離子通道、以及胞內(nèi)和胞外的受體相互作用來改變細(xì)胞的轉(zhuǎn)錄水平以來發(fā)揮抗腫瘤作用。研究方法:本實(shí)驗(yàn)應(yīng)用MTT實(shí)驗(yàn)、流式細(xì)胞術(shù)、細(xì)胞劃痕實(shí)驗(yàn)、Transwell細(xì)胞侵襲實(shí)驗(yàn)檢測了人參皂苷Rg3對骨肉瘤細(xì)胞系MG-63、HOS和U2OS增殖、凋亡、遷移、侵襲的影響和對細(xì)胞周期作用。我們又進(jìn)一步選擇MG-63細(xì)胞作為研究對象,應(yīng)用western blot、定量PCR、流式細(xì)胞術(shù)、小干擾RNA等實(shí)驗(yàn)手段進(jìn)一步探究人參皂苷Rg3誘導(dǎo)細(xì)胞自噬、凋亡以及細(xì)胞周期阻滯的具體作用機(jī)制。實(shí)驗(yàn)結(jié)果:MTT實(shí)驗(yàn)發(fā)現(xiàn)人參皂苷Rg3以劑量依賴的方式抑制三種骨肉瘤細(xì)胞...
