超聲靶向微泡破壞介導(dǎo)BMSCs修復(fù)大鼠急性肝損傷的實驗研究

目的:超聲靶向微泡破壞(Ultrasound targeted microbubble destruction,UTMD)技術(shù)是一種新的藥物和基因遞送方式,靶向特異性強、效率高,已有研究顯示UTMD能增強骨髓間充質(zhì)干細(xì)胞(Bone marrow mesenchymal stem cells,BMSCs)的靶向歸巢率,改善心功能和修復(fù)缺血心肌。本研究中,我們擬探討UTMD對BMSCs歸巢至肝臟的可行性、有效性,進而評估BMSCs對急性肝損傷的治療效果,試圖分析UTMD促進BMSCs靶向歸巢的潛在機制。方法:采用貼壁離心法分離培養(yǎng)大鼠BMSCs,鑒定其增殖特性、三向分化能力和細(xì)胞表面抗原標(biāo)志。攜帶增強型綠色熒光蛋白(Enhanced green fluorescent protein,EGFP)的慢病毒感染BMSCs用于體內(nèi)示蹤。SD大鼠經(jīng)腹腔單次注射1.5g/kg D-氨基半乳糖(D-galactosamine,D-Gal)建立急性肝損傷(Acute liver injury,ALI)模型,檢測24小時后血清學(xué)和肝臟組織結(jié)構(gòu)評價模型是否成功建立。輻照急性肝損傷大鼠,24h后檢測肝組織內(nèi)基...
