【摘要】：正文章簡(jiǎn)介PTPN12是一個(gè)重要的腫瘤抑制因子,并參與調(diào)控很多重要的生理過程。作為磷酸酶,PTPN12有很多底物,但對(duì)其底物的特異性缺乏了解。研究組通過酶學(xué)和結(jié)構(gòu)生物學(xué)方法,發(fā)現(xiàn)了PTPN12催化底物的兩個(gè)重要特征:p Y+1位點(diǎn)的特異性結(jié)合口袋以及由堿性氨基酸為主的活性中心附近識(shí)別區(qū)域。一些特異性位置的氨基酸組合以及結(jié)構(gòu)
[Abstract]:PTPN12 is an important tumor suppressor and is involved in many important physiological processes. As a phosphatase, PTPN 12 has many substrates, but the specificity of its substrate is not well understood. Two important characteristics of PTPN12 catalytic substrate, the specific binding pocket of the 1: pY1 site and the recognition region near the active site dominated by basic amino acids, were found by enzymatic and structural biological methods.
