高住高練對肥胖大鼠脂代謝的影響及機制研究

【摘要】:研究目的:從脂蛋白代謝、脂肪酸代謝(脂肪酸合成、轉(zhuǎn)運、分解、氧化)、脂肪細(xì)胞因子與調(diào)節(jié)激素、PPARα調(diào)控等角度,系統(tǒng)研究高住高練對肥胖大鼠體重和脂代謝的影響及機制。 研究方法:建立肥胖大鼠高住高練0-4周模型,分為0周組、低氧安靜1-4周組、低住低練1-4周組、高住高練1-4周組。水平動物跑臺進(jìn)行耐力訓(xùn)練,訓(xùn)練強度常氧25-26m/min,低氧20-21m/min,1h/d,6d/w,分別持續(xù)1-4周后取材測試。 主要結(jié)果:1)高住高練2-4周較低氧安靜和低住低練顯著降低肥胖大鼠的體重指數(shù),2-4周降低大鼠體重和脂體比優(yōu)于低氧安靜;2)高住高練3、4周顯著降低肥胖大鼠TC, TG,升高高密度脂蛋白,且4周效果優(yōu)于低氧安靜和低住低練;載脂蛋白AⅠ、AⅡ、脂蛋白酯酶升高,載脂蛋白B100、CⅢ降低,參與了脂蛋白代謝的調(diào)節(jié);3)高住高練3周較低住低練顯著降低肥胖大鼠肝臟脂肪酸合成酶基因表達(dá)。高住高練3周顯著增加肥胖大鼠脂肪組織甘油三酯酶基因表達(dá)和激素敏感酯酶含量,且高于低住低練;高住高練4周提高肥胖大鼠腓腸肌肉堿脂酰轉(zhuǎn)移酶、脂肪酸轉(zhuǎn)位酶基因表達(dá)和脂肪LPL與腓腸肌LPL比值,且較低住低練更顯著。高住高練2-4周降低肝臟脂肪酸結(jié)合蛋白和脂肪酸轉(zhuǎn)運蛋白,且低于低住低練;4)高住高練3、4周較低住低練和低氧安靜顯著提高瘦素、瘦素受體、脂聯(lián)素水平,降低內(nèi)酯素水平;5)高住高練3、4周升高胰高血糖素、腎上腺素、生長激素水平,降低胰島素和甲狀腺素水平,且高住高練4周胰島素顯著低于低氧安靜,甲狀腺素顯著低于低住低練,胰高血糖素顯著高于低氧安靜,高住高練3周生長激素高于低氧安靜;6)高住高練組4周顯著性升高大鼠腓腸肌PPARα mRNA水平;3周顯著高于低住低練。 研究結(jié)論:高住高練3、4周較低住低練升高肥胖大鼠PPARα mRNA、瘦素和瘦素受體、脂聯(lián)素、胰高血糖素、腎上腺素、生長激素,降低內(nèi)脂素、胰島素和甲狀腺素,在其調(diào)節(jié)作用下,高住高練3、4周較低住低練改善脂蛋白代謝,抑制脂肪合成,促進(jìn)脂肪分解和氧化,減少脂肪轉(zhuǎn)運至胞內(nèi),導(dǎo)致脂代謝負(fù)平衡,從而減少體脂,降低體重指數(shù)。
[Abstract]:Objective: to study the effects and mechanisms of high altitude training on body weight and lipid metabolism in obese rats from the perspectives of lipoprotein metabolism, fatty acid metabolism (fatty acid synthesis, transport, decomposition, oxidation, adipocyte factor and regulating hormone PPAR 偽). Methods: the obese rats were divided into 0 week group, 1 4 weeks hypoxia quiet group, 1 4 weeks low living and low training group and 1 4 weeks high living high training group. Endurance training was performed on the horizontal animal running platform. The intensity of training was 25-26m / min, hypoxic 20-21m / min, 1h / d / min, 6d / w, respectively, and the samples were tested after 1-4 weeks.
