MoYvh1調(diào)控稻瘟病菌生長發(fā)育及致病力的分子機制研究

稻瘟病菌(Magnaporthe oryzae)引起的稻瘟病是全球稻區(qū)廣泛發(fā)生的、水稻上最重要的毀滅性真菌病害,嚴(yán)重威脅著全球糧食生產(chǎn)安全,其造成的損失可以養(yǎng)活6000萬人口。但是直到現(xiàn)在,由于病菌的快速變異以及在水稻上易產(chǎn)生抗性,傳統(tǒng)的作物耕作方式或者藥劑防治手段都無法控制病菌的擴展與危害。盡管對稻瘟病菌的致病機制進(jìn)行了長期的研究,但是其依舊是包括水稻、大麥、小麥以及小米等禾本科植物上最重要的毀滅性真菌病害。因此,進(jìn)一步深入解析稻瘟病菌的侵染循環(huán)以及與寄主互作中的分子作用機制,對控制病菌危害,開發(fā)針對性藥劑,提供高效低毒的防控策略具有重要的意義。真核生物中蛋白磷酸酶對其具有重要的調(diào)控功能。在釀酒酵母中雙特異性蛋白磷酸酶(DSP)ScYvh1調(diào)控酵母生長、產(chǎn)孢以及糖原積累過程。然而,在絲狀真菌中Yvh1的功能仍不清楚。在本章的研究中,我們鑒定到稻瘟病菌中Yvh1的同源蛋白MoYvh1。通過同源重組敲除MoYVH1后發(fā)現(xiàn),MoYVH1缺失導(dǎo)致病菌營養(yǎng)生長顯著減緩、產(chǎn)孢量降低、致病力減弱。ΔMoyvh1缺失突變體表現(xiàn)出細(xì)胞壁完整性缺陷,同時對氧化脅迫更為敏感。進(jìn)一步研究發(fā)現(xiàn)ΔMoyvh1突...
