鈉鈣交換體亞型1介導(dǎo)CDX2上調(diào)促進(jìn)Barrett食管發(fā)生發(fā)展的研究

目的:Barrett’s食管(Barrett’s Esophagus,BE)是發(fā)生于食管下段鱗柱狀交界區(qū)的鱗狀上皮被單層柱狀上皮所替代的一種病理現(xiàn)象,常伴有腸上皮化生,是一種公認(rèn)的食管腺癌的癌前病變。研究證實(shí)膽汁反流能夠上調(diào)腸型分化標(biāo)志物尾側(cè)型同源盒轉(zhuǎn)錄因子-2(Caudal-related homeobox transcription factor-2,CDX2)的表達(dá),從而促進(jìn)BE的發(fā)生、發(fā)展,但具體機(jī)制不清。我們的前期研究發(fā)現(xiàn)脫氧膽酸(Deoxycholic acid,DCA)刺激正常食管上皮細(xì)胞過程中伴隨著細(xì)胞內(nèi)鈣的增高,并促進(jìn)正常食管上皮細(xì)胞中CDX2的表達(dá)。這一結(jié)果提示細(xì)胞內(nèi)鈣活化可能參與上調(diào)CDX2的表達(dá)。提示參與調(diào)控細(xì)胞內(nèi)鈣活化的離子通道在這一事件中起著舉足輕重的作用。鈣離子是細(xì)胞內(nèi)重要的第二信使,參與了細(xì)胞多種生理與病理過程。鈉鈣交換體亞型1(Na+/Ca2+exchanger1,NCX1)是調(diào)控細(xì)胞內(nèi)外鈣平衡的重要離子通道之一。我們既往的研究發(fā)現(xiàn),NCX1參與調(diào)控肝癌細(xì)胞內(nèi)鈣重構(gòu)并促進(jìn)肝癌細(xì)胞的增殖、分化。G蛋白偶聯(lián)膽汁酸...
