ATF5在鼻咽癌侵襲和轉(zhuǎn)移中的作用及其機制研究

目的:探討轉(zhuǎn)錄激活因子5(Activating transcription factor 5,ATF5)對鼻咽癌細(xì)胞侵襲及遷移能力的影響及其可能分子機制。方法:以人鼻咽癌CNE2Z細(xì)胞為研究對象,采用慢病毒載體shRNA-ATF5干擾技術(shù)沉默CNE2Z細(xì)胞ATF5表達及實時熒光定量聚合酶鏈反應(yīng)(Quantitative Real-time Polymerase Chain Reaction,qRT-PCR)檢測慢病毒shRNA-ATF5載體干擾效率;以未感染慢病毒的CNE2Z細(xì)胞作空白對照(Control組),慢病毒空載體LV-shRNA-NC感染CNE2Z細(xì)胞作為陰性對照(NC組),慢病毒載體shRNA-ATF5(LV-shRNA-ATF5)感染CNE2Z細(xì)胞為實驗組(shRNA-ATF5組);再運用噻唑藍(lán)(Methylthiazolyl Blue Tetrazolium,MTT)法及克隆形成實驗檢測CNE2Z細(xì)胞增殖能力,劃痕實驗、Transwell遷移實驗、侵襲實驗檢測CNE2Z細(xì)胞遷移及侵襲能力;采用Affymetrix人類基因表達譜芯片技術(shù)比較感染shRNA-ATF5慢病毒后...
