【摘要】：目的通過研究AE患者外周血中DC的成熟度,探究DC在AE免疫發(fā)病機制中的作用并分析其及與AE的發(fā)生發(fā)展的關(guān)系。方法1選取2015年9月至2016年6月于青海大學(xué)附屬醫(yī)院肝膽胰二科初診并經(jīng)病理證實為AE患者及隨機抽取同期年齡相仿健康體檢者各20例,采集外周空腹抗凝全血2ml,流式細胞儀檢測外周血CD80~+細胞、CD86+細胞、CD83~+細胞及(CD80~++CD86++CD80~+)/CD14+比值。2選取2016年6月到2017年2月于青海大學(xué)附屬醫(yī)院肝膽胰二科初診并經(jīng)病理證實為AE患者及隨機抽取同期年齡相仿健康體檢者30例,加入細胞因子GM-CSF,IL-4及TNF-α進行體外培養(yǎng)誘導(dǎo),獲取大量DC,利用流式細胞儀檢測DC表面CD83、CD86、CD80表達率。結(jié)果1 AE組與對照組外周血檢測DC表面的共刺激分子(CD80~+CD83~+CD86)/CD14無統(tǒng)計學(xué)差異(P0.05),CD86細胞表達較對照組低(P0.05),且表達有統(tǒng)計學(xué)意義。2 AE組外周血單核源性培養(yǎng)DC表面的共刺激分子CD80、CD83、CD86的表達水平(41.71±8.16、39.69±8.05、50.79±7.72)均低于對照組CD80、CD83、CD86的表達水平(62.27±5.94、56.37±7.33、72.00±7.36),差異均有統(tǒng)計學(xué)意義(P0.05)。結(jié)論本研究顯示AE患者外周血DC成熟度比對照組低,存在成熟障礙,可能會導(dǎo)致宿主抗原提呈功能下降,機體抗感染能力降低,從而促進了多房棘球蚴感染的進展。
[Abstract]:Objective to study the maturity of DC in peripheral blood of patients with AE, to explore the role of DC in the pathogenesis of AE and to analyze the relationship between DC and the occurrence and development of AE. Methods from September 2015 to June 2016, 20 patients with AE and 20 healthy people of the same age were selected from the second Department of Hepatopancreatology, affiliated Hospital of Qinghai University, from September 2015 to June 2016. The peripheral fasting anticoagulant whole blood (2ml) was collected. CD80~ cells and CD86 cells in peripheral blood were detected by flow cytometry. CD83~ cells and (CD80~ CD86 CD80~) / CD14 ratio were selected from June 2016 to February 2017 in the second Department of liver, Bile and pancreas, affiliated Hospital of Qinghai University and pathologically proved to be AE patients, and 30 healthy persons of similar age were randomly selected for the same period. A large number of DC, was obtained by the addition of cytokines GM-CSF,IL-4 and TNF- 偽 in vitro. The expression rate of CD83,CD86,CD80 on DC was detected by flow cytometry. Results 1 there was no significant difference in CD80~ CD83~ CD86 / CD14 on DC surface between AE group and control group (P0.05), but the expression of CD86 cells was lower in AE group than in control group (P0.05). 2 the expression of CD80,CD83,CD86 on the surface of mononuclear culture DC in AE group was significantly lower than that in control group (41.71 鹵8.1659.69 鹵8.05 鹵50.79 鹵7.72). The expression level of CD86 (62.27 鹵5.94) was 56.37 鹵7.33 鹵72.00 鹵7.36, the difference was statistically significant (P0.05). Conclusion this study shows that the maturity of peripheral blood DC in patients with AE is lower than that in the control group, which may lead to the decline of host antigen presentation function and the decrease of body anti-infection ability, thus promoting the progress of infection of echinococcus multilocularis.
