石蒜科植物朱頂紅中生物堿類成分的分離與結(jié)構(gòu)鑒定

【摘要】:朱頂紅Hippeastrum vittatum(Ait.)Herb為石蒜科(Amaryllidaceae)朱頂紅屬(Hippeastrum),多年生草本植物,主要為栽培品種,其鱗莖入藥,味辛,性溫,有小毒,具有解毒消腫的功效,主治癰瘡腫毒。本論文對(duì)以下三個(gè)方面進(jìn)行了研究:1、本文對(duì)朱頂紅及其同屬植物,對(duì)其化學(xué)成分和藥理作用等方面的研究進(jìn)展進(jìn)行了綜述。2、對(duì)朱頂紅鱗莖中的生物堿類成分進(jìn)行了提取分離:取朱頂紅鱗莖采用甲醇浸提后,減壓回收甲醇,將濃縮后的水溶液經(jīng)D-101大孔樹脂柱洗脫,收集70%甲醇洗脫部分,減壓回收甲醇,得到朱頂紅鱗莖中的生物堿類粗提取物。之后采用正相硅膠柱色譜法,分離得到9個(gè)生物堿類成分:I、II、III、IV、V、VI、VI I、VIII、IX。3、通過(guò)理化性質(zhì)測(cè)定及現(xiàn)代波譜學(xué)方法,對(duì)分離得到的生物堿類成分進(jìn)行了結(jié)構(gòu)鑒定,分別為:山小星蒜堿(montanine)(I),11-表山小星蒜堿(11-epimontanine)(II),8-O-去甲基高石蒜堿(8-O-demethylhomolycorenine)(III),小星蒜堿(hippeastrine)(IV)、4a-表-小星蒜堿(4a-epi-hippeastrine)(V)、石蒜堿(lycorine)(VI)、偽石蒜堿(pseudolycorine)(VII)、恩其明(ungeremine)(VIII)、8-O-demethylvasconine(IX)。其中:化合物II、V為新化合物,化合物III、VII、VI II、IX為首次從朱頂紅中分離得到。
[Abstract]:Hippeastrum vittatum (Ait.) Herb is a perennial herbaceous plant of the genus (Hippeastrum), of Lycoris (Amaryllidaceae). It is mainly cultivated. Its bulb is used in medicine, taste and Xin, sex is warm, it has small poison, it has the effect of detoxification and detumescence, and it is mainly used to treat carbuncle swelling poison. In this paper, the following three aspects were studied: 1. The advances in chemical composition and pharmacological action were reviewed. 2. The alkaloids in the bulb were extracted and separated: methanol was extracted from the bulb and methanol was recovered at low pressure. The concentrated aqueous solution was eluted on D-101 macroporous resin column, 70% methanol was collected and methanol was recovered at low pressure to obtain crude alkaloid extract from red bulb. After that, nine alkaloids were isolated by normal phase silica gel column chromatography. The structures of the alkaloids were identified by physicochemical properties and modern spectroscopic methods. The results are as follows: (montanine) (I), 11- 11-epimontanine) (II), 8-O- 8-O-demethylhomolycorenine) (III), allicin (hippeastrine) (IV), 4a- 4a-epi-hippeastrine) (V), allicin (lycorine) (VI), pseudoallicin (pseudolycorine) (VII), en Keemin (ungeremine) (VIII), 8-O-demethylvasconine (IX). Among them, compound II,V is a new compound and compound III,VII,VI II,IX is isolated from red for the first time.
