雪菊黃酮有效部位化學(xué)成分與藥材質(zhì)量研究

【摘要】:目的:對(duì)雪菊中黃酮類化合物進(jìn)行分離純化及結(jié)構(gòu)鑒定;研究雪菊黃酮部位和藥材中主要成分的含量,及藥材中主要成分的含量與性狀特征的相關(guān)性;通過對(duì)黃酮類化合物的降糖活性的研究,探尋雪菊藥效物質(zhì)基礎(chǔ),為質(zhì)量標(biāo)準(zhǔn)建立奠定基礎(chǔ)。方法:使用硅膠、SephadexLH-20、RP-18反向硅膠等多種柱色譜與Semi-PHPLC技術(shù),并結(jié)合重結(jié)晶等方法,從雪菊提取物中分離黃酮類化合物,通過1M-NMR、13C-NMR、HMQC、MS等技術(shù)鑒定化合物結(jié)構(gòu);采用HPLC-DAD技術(shù)對(duì)雪菊黃酮部位及藥材中的主要成分進(jìn)行含量測定,并建立雪菊藥材HPLC圖譜,對(duì)10批雪菊樣品HPLC圖譜進(jìn)行相似度評(píng)價(jià);觀察雪菊舌狀花的特征,依據(jù)兩色舌狀花的紅褐色部分的長度比例及顏色進(jìn)行分級(jí);以舌狀花特征等級(jí)與主要成分含量為自變量,進(jìn)行聚類分析,將10批樣品進(jìn)行分組,分析雪菊中主要黃酮類成分的含量與雪菊舌狀花顏色特征的相關(guān)性;以小鼠3T3-L1前脂肪細(xì)胞為研究載體,采用葡萄糖氧化酶法考察雪菊黃酮類成分對(duì)細(xì)胞葡萄糖代謝產(chǎn)生的影響,給藥量設(shè)計(jì)為2.5、5、10μg.ml-1。結(jié)合化學(xué)、性狀與藥理研究,闡明雪菊黃酮類成分與藥材質(zhì)量的相關(guān)性,為質(zhì)量標(biāo)準(zhǔn)建立奠定基礎(chǔ)。結(jié)果:本研究共分離得到12個(gè)化合物,鑒定出其中8個(gè)化合物的結(jié)構(gòu),分別是2S',5',5-三羥基二氫黃酮-7-O-β-D-吡喃葡萄糖苷、3',5'二羥基二氫黃酮-7-O-β-D-吡喃葡萄糖苷、2S-3',5,5',7-四羥基二氫黃酮、2S-3',4',7,8-四羥基二氫黃酮、馬里苷、奧卡寧、紫鉚花素-4'-O-β-D-吡喃葡萄糖苷、槲皮素-7-O-β-D-吡喃葡萄糖苷。對(duì)得到的馬里苷、奧卡寧、2S-3',4',7,8-四羥基二氫黃酮、2S-3',5',5-三羥基二氫黃酮-7-O-β-D-吡喃葡萄糖苷4個(gè)主要成分在雪菊黃酮部位中的含量進(jìn)行測定,測定結(jié)果為2S-3',4',7,8-四羥基二氫黃酮的含量為0.053 71mn·mg-1、2S-3',5',5-三羥基二氫黃酮-7-O-β-D-吡喃葡萄糖苷的含量為0.032 12 mg·ng-1、馬里苷的含量為0.118 8 mg·mg-1、奧卡寧的含量為0.047 37 mn·mg-1,其中馬里苷的含量最高。根據(jù)雪菊藥材HPLC圖譜的相似度分析,發(fā)現(xiàn)市購10批雪菊藥材樣品的HPLC圖譜相似度均≥0.95。性狀研究發(fā)現(xiàn)市購不同批次樣品間舌狀花的性狀存在差異。含量測定結(jié)果表明不同批次樣品的馬里苷與奧卡寧含量差異較大。將雪菊舌狀花紅褐色部分的長度、顏色深淺及馬里苷、奧卡寧含量作為自變量進(jìn)行聚類分析,發(fā)現(xiàn)馬里苷含量與紅褐色部分顏色及面積存在正相關(guān)。藥理研究結(jié)果為雪菊黃酮部位及各單體成分會(huì)對(duì)正常小鼠3T3-L1細(xì)胞的糖代謝產(chǎn)生影響,但與對(duì)照組之間不存在顯著性差異。結(jié)論:雪菊黃酮部位及藥材主要成分為查爾酮和二氫黃酮,其中馬里苷含量最高并與藥材舌狀花的性狀特征存在相關(guān)性,建議作為雪菊質(zhì)量評(píng)價(jià)的指標(biāo)性成分。
[Abstract]:Objective: to study the content of flavonoids and the correlation between the content of flavonoids and the characteristics of characters. Through the study on the hypoglycemic activity of flavonoids, the substance foundation of the medicinal effect of Xueju was explored, which laid a foundation for the establishment of quality standard. Methods: Sephadex LH-20 RP-18 reverse silica gel column chromatography and semi-PHPLC were used to isolate flavonoids from the extract of Fructus asiatica. The structures of the compounds were identified by 1M-NMR-13C-NMR-HMQCMS. HPLC-DAD technique was used to determine the contents of flavonoids and the main components in the medicinal materials. HPLC chromatograms were established to evaluate the similarity of the HPLC profiles of 10 batches of snow chrysanthemum samples, and to observe the characteristics of the tongue flowers of snow chrysanthemum. According to the length ratio and color of the reddish brown part of the two-color glossy flower, the characteristic grade and the main component content of the glossy flower were used as independent variables, and the 10 batches of samples were grouped. The relationship between the content of flavonoids in chrysanthemum and the color characteristics of chrysanthemum tongue flower was analyzed, and the effect of flavonoids on glucose metabolism was investigated by glucose oxidase method using 3T3-L1 preadipocytes in mice as the carrier. The dosage was designed to be 2.5 渭 g 路ml ~ (-1), 10 渭 g 路ml ~ (-1). The correlation between flavonoids and the quality of medicinal materials was clarified by combining chemical, character and pharmacological studies, which laid a foundation for the establishment of quality standard. Results: in this study, 12 compounds were isolated and 8 compounds were identified. The results were as follows: (2) Spline 5H _ (5) -trihydroxy dihydroflavone -7-O- 尾 -Dpyranoside 5 '-dihydroxy dihydroflavone -7-O- 尾 -D- pyranoside (2S-3) -7S-38-tetrahydroxy dihydroxy dihydrogenone, Marinine, Okanine, Violotenin-4- O- 尾 -D- glucopyranoside, Glucopyranoside, and pyrrolidine, respectively, were 2S-378-tetrahydroxy dihydroxy dihydroflavone, Marinine, Okanin, Violanol-4- O- 尾 -D- glucopyranoside, pyrrolycoside. Quercetin -7-O- 尾 -Dpyranoside. The content of the four main components of marinine, Okaninine 2S-3, 4-dihydroflavone, 7-O- 尾 -Dpyranoside, was determined in the flavonoid fraction of Schizea chinensis, and the contents of the four main components were determined in the flavonoid fraction of Cassia chinensis, and the contents of the four main components were determined in the flavonoids of Cassia chinensis, and the contents of the four main components were determined.
