【摘要】：研究背景:荷青花[Hylomecon japonica(Thunb.)Prantl et Kündig]為荷青花屬(Hylomecon)植物,別名雞蛋黃花(東北)、拐棗七、刀豆三七、水菖三七,大葉老鼠七等,屬于多年生早春類短命植物,主要分布于我國黑龍江、吉林、遼寧、陜西、四川、安徽、浙江、湖南、湖北及山西南部等地,在日本、朝鮮及俄羅斯西伯利亞等地也有分布。生于海拔300-1800(2400)米的落葉闊葉林下、雜木林下、林緣、溝邊或者沙地,喜水肥條件較好的腐殖土和適當遮陰的環(huán)境。荷青花多以根莖入藥,具有祛風除濕,舒筋通絡(luò),散瘀消腫,止血鎮(zhèn)痛之效。荷青花常與多種中藥合用,用于治療風寒濕痹、風濕關(guān)節(jié)痛、跌打損傷、勞傷、四肢乏力、胃脘痛、痢疾等疾病。實驗目的:查閱文獻發(fā)現(xiàn),有關(guān)荷青花成分的研究很少,從中分離出的化合物主要為生物堿類,而對其它成分的研究目前尚無報道。荷青花屬中的其他植物主要分離出酚類及萜類,整體研究尚不深入�，F(xiàn)代藥理學研究表明,荷青花屬植物抗菌、抗炎作用顯著。為更好的開發(fā)利用荷青花屬植物荷青花的藥用價值,從而為中藥現(xiàn)代化奠定堅實的基礎(chǔ),本文對荷青花中酚類成分進行了系統(tǒng)的研究。實驗方法:取荷青花全草,用70%的乙醇溶液重復冷浸提4次,每次一周。合并提取液,減壓回收乙醇后合并水層,干燥后用水溶解,過濾除去水不溶性雜質(zhì),上樣于已處理好的D101大孔樹脂,依次用0%、10%、30%、50%、70%、95%的乙醇洗脫,收集30%部分,干燥后用水溶,除去水不溶性雜質(zhì),剩余樣品經(jīng)正相硅膠柱分離得到A、B、C、D四部分,利用大孔樹脂柱層析、正相硅膠柱層析、HPLC半制備等手段分離得到五個單體化合物。實驗結(jié)果:利用上述方法,從荷青花中分離得到4個酚類化合物和1個單萜類化合物,并鑒定其結(jié)構(gòu)分別為:I:香葉木素-7-O-β-D-吡喃木糖基(1→6)-β-D-吡喃葡萄糖苷II:苯甲基-α-L-吡喃阿拉伯糖基(1→6)-β-D-吡喃葡萄糖苷III:苯甲基-β-D-吡喃木糖基(1→6)-β-D-吡喃葡萄糖苷IV:苯乙基-α-L-吡喃阿拉伯糖基(1→6)-β-D-吡喃葡萄糖苷V:3-羰基-6-羥基-紫羅蘭酮實驗結(jié)論:化合物I-IV為荷青花植物中首次得到,V為荷青花屬中首次得到。
[Abstract]:Background: Hylomecon japonica(Thunb.)Prantl et K 眉 ndig is a species of Hylomecon, also known as Hylomecon, also known as Hylomecon, also known as Hylomecon, also known as Hylomecon, also known as Hylomecon, also known as Hylomecon, also known as Hylomecon, also known as Hylomecon, also known as Hylomecon, also known as Hylomecon, also known as Hylomecon. It is often used in the treatment of rheumatism and dampness arthralgia, falling injury, labor injury, limb fatigue, epigastric pain, dysentery and other diseases. Objective: to find out that there are few studies on the components of lotus blue and white flowers, and the compounds isolated from them are mainly alkaloids, but there are no reports on the other constituents. Phenols and terpenoids are mainly isolated from other plants in the genus Salvia. Modern pharmacological studies have shown that the plants of the genus Salvia are antibacterial and have significant anti-inflammatory effects. In order to better develop and utilize the medicinal value of lotus blue and white plants and lay a solid foundation for the modernization of traditional Chinese medicine, the phenolic components in lotus blue and white flowers were systematically studied in this paper. Methods: the whole plant was extracted with 70% ethanol solution for 4 times, one week each time. Combine the extract, decompress and recover the ethanol and merge the water layer, dissolve with water after drying, filter out the water insoluble impurities, apply the sample to the treated D101 macroporous resin, then use 010, 1010, 30 and 30% of ethanol to be eluted, collect 30% of the water, and dissolve with water after drying, After the water insoluble impurity was removed, the remaining samples were separated by normal phase silica gel column to obtain four parts of Agna Bu Con D, and five monomers were separated by macroporous resin column chromatography and HPLC semi-preparation by normal phase silica gel column chromatography. Results: four phenolic compounds and one monoterpenoid compound were isolated from the blue and white flowers by the method mentioned above. Their structures were identified as: 1: I: vanillin-7-O- 尾 -Dpyranoxylycosyl 1 (1)-尾 -Dpyranoside (II): phenylmethyl- 偽 -L-pyrrolidene arabinoside (1)-尾 -glucopyranoside (III): benzomethyl- 尾 -D- pyranoxylycoside (1)-尾 -Dpyranoside (IV-1). Experimental results of phenylethyl- 偽 -L-pyrrolidene arabinyl glycosyl 1 and 尾 -Dpyranoside (V: 3-carbonyl -6-hydroxy-ionone) conclusion: the compound I-IV is the first obtained from the plants of the genus Salvia.
