一枝蒿揮發(fā)油對過敏性鼻炎的作用研究

【摘要】:目的:(1)優(yōu)選一枝蒿揮發(fā)油提取工藝。(2)分析不同產地一枝蒿揮發(fā)油的化學成分,對其種類及含量進行對比研究,優(yōu)選一枝蒿藥材。(3)優(yōu)選一枝蒿揮發(fā)油液體制備工藝。(4)分別建立小鼠耳腫脹模型、小鼠腹腔毛細血管通透性模型研究一枝蒿揮發(fā)油抑制炎癥作用。(5)建立小鼠遲發(fā)型超敏反應模型,探討一枝蒿揮發(fā)油的抗過敏作用。(6)建立過敏性鼻炎(AR)大鼠模型,探討一枝蒿揮發(fā)油對過敏性大鼠模型的治療作用。方法:(1)通過單因素實驗,以出油率為指標,優(yōu)選一枝蒿揮發(fā)油提取工藝。(2)利用水蒸氣蒸餾法提取一枝蒿揮發(fā)油,用歸一化法測定相對含量,GC-MS法對三個不同產地一枝蒿揮發(fā)油的化學成分進行鑒定、分析。(3)以一枝蒿揮發(fā)油的增溶量為指標,優(yōu)選一枝蒿揮發(fā)油液體的處方。(4)建立二甲苯致小鼠耳腫脹、冰醋酸致小鼠腹腔毛細血管通透性實驗的動物模型,觀察一枝蒿揮發(fā)油的炎癥抑制作用。(5)采用2,4-二硝基氯苯建立小鼠遲發(fā)型超敏反應模型,探討一枝蒿揮發(fā)油抗過敏的作用。(6)利用卵清蛋白(OVA)氫氧化鋁混懸液進行全身致敏和鼻部抗原攻擊,建立過敏性鼻炎大鼠模型。采用酶聯(lián)免疫吸附(ELISA)法,測定大鼠血清中免疫球蛋白E(IgE)、特異性IgE(s IgE)及白介素-4(IL-4)的含量,并取鼻粘膜做病理組織學檢查,探討一枝蒿揮發(fā)油對大鼠血清IgE、sIgE、IL-4以及鼻粘膜組織結構、肥大細胞、炎性細胞浸潤等情況的影響。結果:(1)藥材粗粉,以1:8的料液比,浸潤5h,回流提取5h,為一枝蒿揮發(fā)油最佳提取工藝。(2)三個不同產地一枝蒿揮發(fā)油的主要化合物及其含量均有差異,并首次分析出十幾種化合物。(3)一枝蒿揮發(fā)油液體制備優(yōu)選工藝為表面活性劑洛沙姆188,與一枝蒿揮發(fā)油比例為56:1,處方量的其余輔料混合可制得外觀穩(wěn)定的一枝蒿揮發(fā)油液體溶液。(4)一枝蒿揮發(fā)油給藥組二甲苯致小鼠耳腫脹程度與模型組相比較輕,差異有顯著性(P0.05)。(5)一枝蒿揮發(fā)油高劑量組與模型組比較,小鼠腹腔毛細血管通透性降低,差異有顯著性(P0.05)。(6)一枝蒿揮發(fā)油給藥組大鼠較模型組,大鼠鼻部病癥、鼻粘膜病變以及大鼠血清IgE、s Ig E及IL-4含量與模型組相比有所減少,有顯著性差異(P0.05)。結論:(1)提取工藝研究結果可為后續(xù)研究提供理論依據(jù)。(2)在理論上初步闡釋了同一采摘時間不同產地一枝蒿揮發(fā)油化學成分種類及含量的不同之處,對其藥效學研究有參考價值。(3)研究制得的一枝蒿揮發(fā)油液體性質均一、穩(wěn)定。(4)一枝蒿揮發(fā)油對過敏性鼻炎有明顯的治療作用。(5)一枝蒿揮發(fā)油能抑制過敏性鼻炎大鼠IL-4的分泌,減少過敏性鼻炎大鼠血清IgE、s IgE含量,并緩解鼻粘膜的病變。(6)一枝蒿揮發(fā)油對過敏性鼻炎大鼠有一定的醫(yī)治效果。
[Abstract]:Objective to analyze the chemical constituents of volatile oil from Artemisia annua from different habitats, and to compare its species and contents.Preparation technology of volatile oil of Artemisia annua L.) the mouse ear swelling model was established, and the mouse model of celiac capillary permeability was established. The model of delayed type hypersensitivity reaction was established by the inhibition of inflammation by the volatile oil of Artemisia annua L.To study the antiallergic effect of essential oil of Artemisia annua L.) to establish the rat model of allergic rhinitis and to explore the therapeutic effect of volatile oil of Artemisia annua on allergic rat model.Methods (1) A single factor experiment was conducted to optimize the extraction process of the volatile oil from Artemisia annua by using the oil yield as the index. The volatile oil of Artemisia annua was extracted by steam distillation.The chemical constituents of the volatile oil of Artemisia annua from three different habitats were identified by GC-MS, and the solubilization of volatile oil of Artemisia annua was used as the index.To establish an animal model of mice ear swelling induced by xylene and capillaries permeability induced by acetic acid in mice.To observe the inflammatory inhibitory effect of volatile oil of Artemisia annua L. 5) mice model of delayed type hypersensitivity was established by using 2o 4-dinitrochlorobenzene.To study the antiallergic effect of essential oil of Artemisia annua L. 6) to establish allergic rhinitis rat model by using ovalbumin OVA) aluminum hydroxide suspension for systemic sensitization and nasal antigen attack.The effects of mast cells and inflammatory cell infiltration.Results the crude powder was the best extraction technology of volatile oil of Artemisia mongolica for 5 h and refluxing for 5 h. The main compounds and contents of volatile oil of Artemisia annua from three different producing areas were different.It is the first time to analyze more than a dozen compounds. 3) the optimum preparation process of volatile oil liquid of Artemisia annua is as follows: surfactant Losham 188. the ratio of volatile oil of Artemisia annua to Artemisia annua is 56: 1. The stable appearance of Artemisia artemisia can be obtained by mixing the other excipients with the prescription amount.The ear swelling of mice induced by xylene in the volatile oil treatment group was lighter than that in the model group.Compared with the model group, the contents of IgE Ig E and IL-4 in nasal mucosal lesions and serum of rats were significantly lower than those in the model group (P 0.05).Conclusion (1) the results of extraction process can provide theoretical basis for further study. (2) the differences of chemical constituents and contents of volatile oil of Artemisia annua from the same picking time and different producing areas were preliminarily explained in theory.The essential oil of Artemisia annua L. is homogeneous in nature and stable. 4) the volatile oil of Artemisia annua has obvious therapeutic effect on allergic rhinitis. 5) the volatile oil of Artemisia annua can inhibit the secretion of IL-4 in allergic rhinitis rats.The essential oil of Artemisia annua has a certain therapeutic effect on allergic rhinitis rats.
