中藥黃芩及其主要成分抗炎活性的研究

【摘要】:本論文通過(guò)動(dòng)物實(shí)驗(yàn)、細(xì)胞實(shí)驗(yàn)等方法考察黃芩及其主要成分黃芩苷、黃芩素的抗炎效果。首先通過(guò)小鼠足腫脹實(shí)驗(yàn)表明,中藥黃芩具有較好的抗炎效果,可以有效抑制小鼠的足腫脹;體外細(xì)胞水平抗炎實(shí)驗(yàn)表明,RAW264.7細(xì)胞模型較NIH3T3細(xì)胞模型更為穩(wěn)定,平行效果也更好。以RAW264.7細(xì)胞為模型,PGE2及NO水平,中藥提取物的抗炎效果在中藥提取物濃度為1mg/ml時(shí),中藥黃芩有較好的抗炎效果,與動(dòng)物實(shí)驗(yàn)抗炎效果數(shù)據(jù)基本一致;體外細(xì)胞水平抗炎實(shí)驗(yàn)表明,采用NIH3T3細(xì)胞檢測(cè)PGE2濃度計(jì)算藥物抗炎效果,黃芩中主要成分抗炎效果最好的為黃芩素,在其濃度"g0.5mg/ml時(shí),其抗炎效果高于陽(yáng)性藥物羅非昔布的抗炎效果;黃芩苷在其濃度為2mg/ml時(shí)與陽(yáng)性藥物的抗炎效果相當(dāng),與動(dòng)物實(shí)驗(yàn)及細(xì)胞水平中藥提取物抗炎實(shí)驗(yàn)結(jié)果一致。采用RAW264.7細(xì)胞檢測(cè)NO濃度計(jì)算藥物抗炎效果,與NIH3T3細(xì)胞檢測(cè)數(shù)據(jù)一致。Western blotting結(jié)果顯示,黃芩提取物刺激后,胞漿中P65表達(dá)明顯減少,,P65的磷酸化水平也有所減低,說(shuō)明所用黃芩提取物具有良好的抗炎效果,與動(dòng)物實(shí)驗(yàn)及細(xì)胞實(shí)驗(yàn)抗炎效果數(shù)據(jù)一致。
[Abstract]:In this paper, the anti-inflammatory effects of Scutellaria baicalensis and its main components baicalin and baicalin were investigated by animal and cell experiments. In vitro cell level anti-inflammatory experiments showed that the RAW264.7 cell model was more stable than the NIH3T3 cell model, and the parallel effect was better. The RAW264.7 cells were used as the model to model the levels of PGE2 and no. The anti-inflammatory effect of traditional Chinese medicine extract at the concentration of 1mg / ml of traditional Chinese medicine extract, Scutellaria baicalensis has a good anti-inflammatory effect, which is basically consistent with the data of anti-inflammatory effect of animal experiment, and the cell level anti-inflammatory experiment in vitro shows that the anti-inflammatory effect of Scutellaria baicalensis L. The anti-inflammatory effect of Scutellaria baicalensis was calculated by NIH3T3 cell detection of PGE2 concentration. The anti-inflammatory effect of baicalin was better than that of rofecoxib at the concentration of "g0.5mg / ml" in Scutellaria baicalensis. Baicalin had the same anti-inflammatory effect as the positive drug at the concentration of 2mg / ml, which was consistent with the results of the animal experiment and the anti-inflammatory test of the Chinese herbal extract at the cell level. The anti-inflammatory effect of the drug was calculated by RAW264.7 cell detection of no concentration. The results of Western blotting showed that the expression of p65 in the cytoplasm of Scutellaria baicalensis was significantly decreased after stimulation with NIH3T3 cells, and the phosphorylation level of P65 was also decreased, which indicated that the extract of Scutellaria baicalensis had a good anti-inflammatory effect. It is consistent with the data of anti-inflammatory effect in animal experiment and cell experiment.
